Osteopontin/OPN (1B20) was detected in immersion fixed HepG2 human hepatocellular carcinoma cell line using Rat anti- Osteopontin/OPN (1B20) Protein G Purified Monoclonal Antibody conjugated to Alexa Fluor® 488 (Catalog # NB110-89062AF488) (green) at 10 µg/mL overnight at 4C. Cells were counterstained with DAPI (dark blue). Cells were imaged using a 100X objective and digitally deconvolved.

The cells were incubated with TGN46 Antibody conjugated to Alexa Fluor 488 (NBP1-49643AF488) at 2 ug/ml for 1 hour at room temperature. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Immunofluorescence microscopy of incoming virus particles, 20 minutes.(A, B) A549 cells were plated onto glass coverslips and the following day replaced with media containing NB-DNJ (200μM) or left untreated. Immunofluorescence microscopy of rVSV-SFTSV particles following UGCG knockdown.(A-D) U-2 OS cells were transfected with siRNAs targeting UGCG (siUGCG) or a non-targeting control (siNegCtrl) and plated onto glass coverslips. Immunocytochemistry/Immunofluorescence: Actin Antibody (mAbGEa) [NB100-74340] - NIH-3T3 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton X-100. The cells were incubated with anti-Actin (mAbGEa) at 5 ug/mL overnight at 4C and detected with an anti-Mouse IgM DyLight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Flow (Intracellular): Actin Antibody (mAbGEa) [NB100-74340] - An intracellular stain was performed on A549 cells with NB100-74340 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by mouse IgM Alexa Fluor 488-conjugated secondary antibody.

(B) Deneddylation test in a heterologous yeast system. A. nidulans DenA can remove Rub1 from CulD in heterologous expression experiments in S. cerevisiae. DenA expressed as native protein or C-terminally fused w/ a V5/His6 epitope tag. Both variants driven by the inducible GAL1 promoter. CulD, N-terminal-fused w/ the LexA activation domain, expressed under control of the constitutive ADH promoter. A. nidulans proteins expressed in S. cerevisiae wild type & delta csn5 background. Western analysis w/ antibodies against Rub1 ( alpha -Rub1), the LexA epitope ( alpha -LexA) & the V5 epitope ( alpha -V5) performed. Detection w/ alpha -Rub1 generated two additional signals upon culD expression, representing LexA-CulD & a second CulD pool where LexA unspecifically cleaved off. Both signals disappeared upon co-expression of DenA indicating deneddylation activity (red arrows). Y27632 (Y10; 10 μM) & calyculin A (A50; 50 μg/ml) were used as negative & positive controls. Western blot analysis & staining with anti-phospho-MLC, anti-total-MLC, & anti-beta actin antibodies was then performed. Phospho-MLC, total-MLC, & beta actin were all obtained with their appropriate protein size bands. Role of NF-kappa B in glutamate-induced increase in the COX-2 protein level in STEP-deficient neurons.A and B, neuronal cultures from (A) WT and (B) STEP KO mice were treated with 50 μM glutamate (Glu) for 20 min and then maintained in their original medium for the specified times (post-Glu time). <b>Genetic Strategies Validation. </b>Simple Western: Ki67/MKI67 Antibody (8D5) [NBP2-22112] - Detection of Ki67/MKI67 by Simple WesternTM. Simple Western lane view shows lysates of HeLa parental cell line and Ki67 knockout (KO) HeLa cell line. A specific band was detected for Ki67/MKI67 at approximately 317 kDa (as indicated) in the parental cell line, but is not detectable in the knockout HeLa cell line using 1:100 of Mouse Anti-Ki67/MKI67 Monoclonal Antibody (Catalog # NBP2-22112). GAPDH is shown as a loading control. This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Immunocytochemistry/Immunofluorescence: Ki67/MKI67 Antibody (8D5) [NBP2-22112] - NIH3T3 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti- NBP2-22112 at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Immunocytochemistry/Immunofluorescence: Ki67/MKI67 Antibody (8D5) [NBP2-22112] - A431 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-Ki-67 (8D5) conjugated to Alexa Fluor 488 [NBP2-22112AF488] at 10ug/ml for 1 hour at room temperature. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunocytochemistry/Immunofluorescence: Ki67/MKI67 Antibody (8D5) [NBP2-22112] - Ki67 antibody was tested at 1:100 in HeLa cells with DyLight 488 (green). Nuclei and alpha-tubulin were counterstained with DAPI (blue) and DyLight 550 (red). Image objective 40x.

Ki67/MKI67 (8D5) was detected in immersion fixed A431 human skin carcinoma cell line using Mouse anti-Ki67/MKI67 (8D5) Protein-G purified Monoclonal Antibody conjugated to Alexa Fluor® 488 (Catalog # NBP2-22112AF488) (green) at 5 µg/mL overnight at 4C. Cells were counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.

RPTECs trigger CD4+ T-cells to proliferate, anoxia-reoxygenation increases proliferation further, whereas CD4+ T-cell differentiation to regulatory T-cells does not occur. CD4+ T-cells were cultured alone or co-cultured with RPTECs for 7 days. RPTECs were previously subjected or not to 90 min of anoxia. Four such experiments were performed. The Western blotting results were normalized for beta -actin and then depicted after normalization for the control group. Albumin overload induces cellular senescence and EMT. Panel (A) depicts the results of one out of three performed experiments. Immunohistochemistry-Paraffin: MMP-9 Antibody (4A3) [NBP2-13173] - Analysis of a FFPE tissue section of human esophageal cancer using MMP9 antibody (clone 4A3) at 1:200. The staining was developed with HRP-labelled secondary antibody and DAB reagent followed by hematoxylin counterstaining. This antibody generated a specific extracellular and cytoplasmic staining primarily in the cancer cells while the signal was pretty weak in the tumor stroma.

Immunohistochemistry-Paraffin: MMP-9 Antibody (4A3) [NBP2-13173] - Analysis of a FFPE section of human esophageal cancer using MMP9 antibody (clone 4A3) at 1:200. The staining was developed with HRP-labelled secondary antibody and DAB reagent followed by hematoxylin counterstaining. This antibody clone generated an extracellular and cytoplasmic staining mainly in the cancer cells while the signal was very weak in the stromal cells of the tumor.

Western Blot: MMP-9 Antibody (4A3) [NBP2-13173] - 2-DZ analysis & identification of MMP-9 by Western blot. Left panels (A & B): 2-D zymography (2-DZ) of serum samples from healthy control (A) & from an inactive RR-MS patient not subjected to therapy (same patient as in Fig. 2; B). For 2-DZ, aliquots of 35 μl of serum were resuspended in the rehydration solution & subjected to isoelectrofocusing (IEF; 1st dimension) on IPG Dry-Strips of 13 cm in a linear pH gradient of 4–7. After IEF, IPG strips were equilibrated & then applied for the 2nd dimension in a 8.5% (w/v) polyacrylamide gel copolymerized with 0.1% (w/v) gelatin. The isoforms & charge variants of MMP-2 & MMP-9 appear as clear spots of digestion on the dark background of the gel. Right panels (A' & B') represent Western blot analysis of the same sera shown in A & B. Aliquots of 35 μl of serum samples (instead of the usual 20 μl) were subjected to 2D electrophoresis (2-DE) by using 8.5% (w/v) polyacrylamide gels without gelatin. After transfer of the proteins, the nitrocellulose membranes were incubated with an anti MMP-9 (Ab-8) Mouse mAb (IA5) at concentration of 2.66 μg/ml. Simple Western: EAAT1/GLAST-1/SLC1A3 Antibody - BSA Free [NB100-1869] - Protein changes in ONHAs corroborate bioenergetics data. (A) Glucose transporter-1 protein levels in Stretched ONH astrocytes are significantly higher than Control (*p = 0.0225, n = 7 Control, n = 8 Stretch). Retinal lysate from a 2 month-old mouse was used as a positive control for each protein analyzed, while negative control was the signal obtained when no primary antibody was included in the capillary. (B) Lactate dehydrogenase-A, the astrocyte-specific isoform of the enzyme that catalyzes the interconversion of pyruvate & lactate, has equivalent protein levels in Control & Stretch ONH astrocytes. Human astrocytes express MHC-II in brain tissue from PD patients and healthy controls. The alpha SYN pathology was confirmed in the PD cases by immunostaining of alpha SYN and phosphorylated alpha SYN (p-alpha SYN) (a). The PD brains express higher levels of MHC-II compared to control brains (b). MHC-II levels correlate with the expression of p-alpha SYN in PD brains (c). There is a correlation between the MHC-II levels in the mesencephalon and striatum in the PD brains, but not in the control brains (d, e). Astrocytes labeled with GLAST (f) or GFAP (g) are expressing high levels of MHC-II, as well as Iba1-labeled microglia (h). Quantifications of the immunostainings revealed that 31% of the MHC-II area overlapped with the astrocyte marker GLAST (i), 15% overlapped with the reactive astrocyte marker GFAP (j), and 32 % overlapped with the microglia marker Iba1 in the PD brain (k). A similar overlap between MHC-II and GLAST, GFAP, and Iba1 (35%, 10%, and 33%, respectively) was revealed from the analysis of the control brains (j–k). Western analysis of cell lysates.a Western analysis of primary neurons, astrocytes, microglia, and OPCs derived from WT and PINK1 KO rats. Cultured cells were treated with 100 nM valinomycin or DMSO control for 4 h at 37 C, then harvested for western analysis. Ten micrograms of cell lysate protein was loaded in each lane, resolved by SDS-PAGE, transferred to PVDF membrane, and blotted using anti-pS65-Ub antibody. The same membrane was re-probed using the antibodies indicated on the lower panels as markers for neurons ( beta 3-tubulin), astrocytes (ALDH1L1), microglia (Iba-1), and OPCs (Olig-2). b Mean +/− SEM pS65-Ub level measured by densitometry of western blot membranes from four independent experiments.
