Western Blot: mCherry Antibody [NBP2-25158] - Analysis of HEK293 cell lysates, and recombinant protein solutions using chicken mCherry pAb, dilution 1:2000 (Green). [1] protein standard, [2] HEK293, [3] HEK293 cells transfected with an mCherry-HA constract, [4] pure recombinant mCherry protein (predicted molecular weight ~ 27kDa), [5] pure recombinant GFP protein and [6] HEK293 transfected with a GFP construct. The major band at about 30kDa corresponds to mCherry-HA protein and the slightly larger recombinant form runs at about 33kDa due to presence of a His tag and other vector derived sequence. The mCherry antibody reacts strongly with both mCherry constructs and does not react with GFP protein. The blot was simultaneously probed with mouse HSP60 mAb, dilution 1:10000 (Red), which reveals a band at 60kDa only in the cell lysates.

Western blot analysis of HEK293 cell lysates using chicken pAb to mCherry, NBP2-25158, dilution 1:2,000, in green, and rabbit pAb to GAPDH, RPCA-GAPDH, dilution 1:5,000, in red: [1] protein molecular weight standard, [2] untransfected HEK293 control cells, [3] HEK293 cells transfected with pCI-Neo-mod vector expressing two tdTomato protein domains, [4] HEK293 cells transfected with pCI-Neo-mod vector expressing one mCherry-HA protein domain, and [5] HEK293 cells transfected with pCI-Neo-mod vector expressing one GFP domain. NBP2-25158 recognizes tdTomato and mCherry proteins revealing major bands at about 60kDa and 30kDa, but does not recognize GFP. The red band at 37kDa corresponds to GAPDH protein here used as a loading control.

Western blot shows lysates of human heart tissue and iBJ6 human induced pluripotent stem cell line undifferentiated or cardiac differentiated. PVDF membrane was probed with 0.25 ug/mL of Mouse Anti-alpha-Smooth Muscle Actin Monoclonal Antibody (Catalog # NBP2-33006) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). The cells were incubated with anti-Fibronectin at 2 ug/mL overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at 1:500. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at 1:1000 and detected with an anti-mouse DyLight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Flow (Intracellular): NQO-1 Antibody (A180) [NB200-209] - An intracellular stain was performed on U87MG cells with NQO1 (A180) antibody NBP2-24917 (blue) along with a matched isotype control NBP2-27287 (orange). Cells were fixed with 4% PFA and permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by mouse F(ab)2 IgG (H+L) APC-conjugated secondary antibody (F0101B, R&D Systems).

Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Western Blot: GRP78/HSPA5 Antibody [NBP1-06274] - Analysis of MAM-associated protein levels in control and patients' fibroblasts.(A) Equal amounts from one control and the five patients were loaded (50 ug of total cell lysates) and subjected to Western Blot with anti-Grp75, anti-sigma-1R and anti-Mfn2 antibodies. We used anti-GAPDH antibody to ensure equal amounts of protein loaded in each lane. This result is representative of three independent experiments. Immunocytochemistry/Immunofluorescence: GRP78/HSPA5 Antibody [NBP1-06274] - NIH-3T3 cells were fixed and permeabilized for 10 minutes using -20C MeOH. The cells were incubated with anti-GRP78/HSPA5 at 2 ug/mL overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Western Blot: GRP78/HSPA5 Antibody [NBP1-06274] - Total protein from human HeLa and A431 cells, mouse 3T3 cells and rat PC12 cells was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 1.0 ug/mL anti-GPR78 in blocking buffer and detected with an anti-rabbit HRP secondary antibody using chemiluminescence.

Immunocytochemistry/Immunofluorescence: GRP78/HSPA5 Antibody [NBP1-06274] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton-X100. The cells were incubated with anti-GRP78/HSPA5 at 5 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Flow Cytometry: GRP78/HSPA5 Antibody [NBP1-06274] - An intracellular stain was performed on NIH3T3 cells with GRP78/HSPA5 Antibody NBP1-06274AF488 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 488.

Flow Cytometry: GRP78/HSPA5 Antibody [NBP1-06274] - Analysis using Alexa Fluor (R) 488 conjugate of NBP1-06274. An intracellular stain was performed on HeLa cells with GPR78/HSPA5 antibody NBP1-06274 (blue) and a matched isotype control NBP2-24893 (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. 1 ug of antibody was added to 100 uL of staining buffer and cells were incubated for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 488.

Flow Cytometry: GRP78/HSPA5 Antibody [NBP1-06274] - An intracellular stain was performed on Jurkat cells with Alexa Fluor 647 conjugate of GPR78/HSPA5 antibody NBP1-06274AF647 (blue) and a matched isotype control NBP2-24893AF647 (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 647.

Flow Cytometry: GRP78/HSPA5 Antibody [NBP1-06274] - An intracellular stain was performed on HeLa with NBP1-06274 and a matched isotype control. Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550.

Immunohistochemistry-Paraffin: Collagen IV Antibody [NB120-6586] - Collagen IV antibody at 1:400, 45 min RT showed strong staining in FFPE sections of human kidney (left) with strong red staining observed in glomeruli and liver (right) with strong staining in sinusoids. Staining for both tissues was consistent with a basement membrane distribution. Slides were steamed in 0.01 M sodium citrate buffer, pH 6.0 at 100C for 20 minutes for antigen retrieval.

Cells were stained with the NorthernLights(TM) 557-conjugated Donkey anti-Rabbit IgG Secondary Antibody [NL557] (red; Catalog # NL004) and counterstained with DAPI (blue).
