This is specific for HIF-1 beta/ARNT. It is not known if NB100-110 cross-reacts with ARNT2 which is related to HIF-1 beta/ARNT but is the product of a different gene.

Application Notes
This HIF-1 beta antibody is useful for Western blot, Immunoprecipitation, Chromatin Immunoprecipitation and Immunohistochemistry on paraffin-embedded sections. This antibody did not work in Immunocytochemistry. By Western blot, this antibody recognizes a band at ~92 kDa representing HIF-1 beta in induced tissues and cells. Background non-specific bands are minimal on MCF7 human nuclear extracts. Background may increase with whole cell extract preparations. Lower dilutions may be needed for whole cell extract preps. Use in Immunocytochemistry/immunofluorescence reported in scientific literature (PMID 17003483). Use in Gel supper shift assay reported in scientific literature (PMID 11046109).In Simple Western only 10-15 uL of the recommended dilution is used per data point.

HIF-1 is a heterodimer composed of HIF-1 alpha and HIF-1 beta subunits. Both subunits are constantly translated. However, under normoxic conditions, human HIF-1 alpha is hydroxylated at Pro402 or Pro564 by a set of HIF prolyl hydroxylases, is polyubiquinated, and eventually degraded in proteosomes. Under hypoxic conditions, the lack of hydroxylation prevents HIF degradation and increases transcriptional activity. Therefore, the concentration of HIF-1 alpha increases in the cell. In contrast, HIF-1 beta remains stable under either condition. HIF-1 beta is a series of aryl hydrocarbon receptor nuclear translocator (ARNT) gene products.

  1. So this NB100-110 does not have a concentration. How much would you recommend to use per chip reaction where I normally use 5 to 10 ug of antibody?
    • Our HIF-1 beta product NB100-110 is provided as whole antisera, so we have not determined the protein concentration. We recommend using a 1:10-1:500 dilution for ChIP. You will need to determine the optimal amount for your experiment.
