Flow Cytometry: Noxa Antibody (114C307.1R) [NBP3-07012] - An intracellular stain was performed on Ntera2 cells with Noxa Antibody [114C307.1R] NBP3-07012 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1.0 ug/mL for 30 minutes at room temperature, followed by Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (35503, Thermo Fisher).

Immunocytochemistry/Immunofluorescence: Noxa Antibody (114C307.1R) [NBP3-07012] - HeLa cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.05% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-Noxa Antibody (114C307.1R) NBP3-07012 at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Immunohistochemistry-Paraffin: Noxa Antibody (114C307.1R) - BSA Free [NBP3-07012] - Immunohistochemical analysis of bouin-fixed, paraffin-embedded chronic lymphocytic leukemia (CLL) xenograft stained with Noxa antibody (1:2000), peroxidase-conjugate and DAB chromogen. Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 5 ug/mL for 30 minutes at room temperature. Both antibodies were directly conjugated to Alexa Fluor 700.

Western Blot: mCherry Antibody [NBP2-25158] - Analysis of HEK293 cell lysates, and recombinant protein solutions using chicken mCherry pAb, dilution 1:2000 (Green). [1] protein standard, [2] HEK293, [3] HEK293 cells transfected with an mCherry-HA constract, [4] pure recombinant mCherry protein (predicted molecular weight ~ 27kDa), [5] pure recombinant GFP protein and [6] HEK293 transfected with a GFP construct. The major band at about 30kDa corresponds to mCherry-HA protein and the slightly larger recombinant form runs at about 33kDa due to presence of a His tag and other vector derived sequence. The mCherry antibody reacts strongly with both mCherry constructs and does not react with GFP protein. The blot was simultaneously probed with mouse HSP60 mAb, dilution 1:10000 (Red), which reveals a band at 60kDa only in the cell lysates.

Western blot analysis of HEK293 cell lysates using chicken pAb to mCherry, NBP2-25158, dilution 1:2,000, in green, and rabbit pAb to GAPDH, RPCA-GAPDH, dilution 1:5,000, in red: [1] protein molecular weight standard, [2] untransfected HEK293 control cells, [3] HEK293 cells transfected with pCI-Neo-mod vector expressing two tdTomato protein domains, [4] HEK293 cells transfected with pCI-Neo-mod vector expressing one mCherry-HA protein domain, and [5] HEK293 cells transfected with pCI-Neo-mod vector expressing one GFP domain. Immunocytochemistry/Immunofluorescence: Fibronectin Antibody - BSA Free [NBP1-91258] - NIH-3T3 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-Fibronectin at 2 ug/mL overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at 1:500. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at 1:1000 and detected with an anti-mouse DyLight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Fibronectin was detected in immersion fixed HepG2 human hepatocellular carcinoma cell line using Rabbit anti-Fibronectin Affinity Purified Polyclonal Antibody conjugated to Alexa Fluor® 647 (Catalog # NBP1-91258AF647) (light blue) at 10 µg/mL overnight at 4C. The expression of Nrf2 & NQO1 was detected by western blotting. Western Blot: NQO-1 Antibody (A180) [NB200-209] - S-(-)equol primarily up-regulated Nrf2 protein expression activating ARE-dependent response.(A) Whole EA.hy926 cells lysates of cells treated with 250 nM S-(-)equol, 500 nM daidzein, or 50 µM tBHQ for 24 h were subjected to immunoblot analysis with anti-Nrf2, anti-HO-1, anti-NQO1, & anti-beta -tubulin antibodies. (B) Total HUVECs lysates of cell treated with 250 nM S-(-)equol for 24 h were immunoblotted with anti-Nrf2 & anti-beta -tubulin antibodies. NQO-1 (A180) was detected in immersion fixed U-2 OS human osteosarcoma cell line using Mouse anti-NQO-1 (A180) Protein G Purified Monoclonal Antibody conjugated to DyLight 650 (Catalog # NB200-209C) (light blue) at 10 µg/mL overnight at 4C. Cells were counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.

Analysis of immune cells infiltration in cerebellum was performed at 4 weeks by immunohistochemistry using antibodies against (A) CD4, a marker of T lymphocytes (F(3,8) = 4.511, p < 0.01) & (B) IBA1, a marker of meningeal & perivascular macrophages (K-W(4,10) = 27.76, p < 0.0001). Rats with mild liver damage show immune cells infiltration in cerebellum at 4 weeks, which is prevented by rifaximin. Immunocytochemistry/Immunofluorescence: PCNA Antibody (PC10) [NB500-106] - PC12 cells were fixed and permeabilized for 10 minutes using -20C MeOH. The cells were incubated with anti-PCNA Antibody (PC10) NB500-106 at 1 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Immunocytochemistry/Immunofluorescence: PCNA Antibody (PC10) [NB500-106] - HeLa cells were fixed and permeabilized for 10 minutes using cold (-20C) MeOH. The cells were incubated with anti-PCNA [PC10] at 5 ug/mL overnight at 4C and detected with an anti-mouse IgG Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Flow Cytometry: PCNA Antibody (PC10) [NB500-106] - An intracellular stain was performed on HepG2 cells with PCNA Antibody (PC10) NB500-106G (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 10 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to DyLight 488.

Western Blot: PCNA Antibody (PC10) [NB500-106] - HeLa cell nuclear extract was resolved by electrophoresis, transferred to nitrocellulose and probed with monoclonal anti-PCNA antibody. Proteins were visualized using a goat anti-mouse secondary conjugated to HRP and a chemiluminescence detection system.

Western Blot: PCNA Antibody (PC10) [NB500-106] - Whole cell protein from human HeLa, A431, mouse Neuro2A and rat PC12 cells was separated on a 4-20% gel by SDS-PAGE, transferred to 0.2 um PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 1.0 ug/mL anti-Histone h3 in block buffer and detected with an anti-mouse HRP secondary antibody using chemiluminescence.
