Application Notes
This Nanog antibody is useful for Western Blot, Immunocytochemistry/Immunofluorescence, Immunoprecipitation, and Immunohistochemistry on paraffin embedded tissue. Prior to immunostaining paraffin tissues, antigen retrieval with Tris-EDTA (pH 9.0) is recommended. Use in Chromatin Immunoprecipitation and Flow Cytometry was reported in a scientific publication (PMID: 23735977).

In stem cell development, the influence of the transcription factor Nanog is important for appropriate differentiation, or conversely to maintain their pluripotent state. Islet survivor cells, generated for progenitor cell testing, have nuclear expression of OCT4, SOX2 and Nanog, indicating a probability that they can maintain pluripotent characteristics in experimentation. In brain cancer, there are some stem cell like behaviors, and the cancer cells exhibiting these characteristics demonstrate high concentrations of Nanog, influenced by p53. Nanog seems to be very important for stem cell growth, maintenance and differentiation, but is usually co-expressed and co-activated by other factors, indicating the need for further mechanistic exploration into its activity.

Nanog antibody was used for IHC-P application on sections of tail skin from tissue specific doxycycline-inducible Nanog transgenic/TG mouse model (K5-rt-TA^tg/.;Col1a1^tetO-Nanog/?+?) and their control/CTR littermates lacking the rtTA transactivator (Col1a1^tetO-Nanog/?+) treated with 2?mg/ml of doxycycline/DOX for 48?hours (Figure 1C), and on CTR / TG papillomas (Figure S2E, Figure S3A). IHC-P was also performed on tail skin of TPA-treated CTR and TG mice (Figure 1F), and on SCC and lymph node metastasis from the same mouse (Figure S2F).
