![Western Blot: EGLN3/PHD3 Antibody - BSA Free [NB100-139] - 5-dAzaC effect on PHD3 transcript (A) & protein (B) levels in HCT116 & DLD-1 CRC cells. HCT116 & DLD-1 cells were cultured in DMEM for 6, 24 & 48 h either in the absence or in the presence of 5-dAzaC at a concentration of 1.00 or 5.00 μM under hypoxic or normoxic conditions. After incubation the cells were used for total RNA isolation & protein isolation. Total RNA was reverse-transcribed, & PHD3 cDNA levels were determined by RQ-PCR relative quantification analysis. RQ-PCR results were standardized by the geometric mean of PBGD & hMRPL19 cDNA levels. PHD3 cDNA levels are expressed as a multiplicity of the respective controls. ![Immunohistochemistry-Paraffin: p19ARF/CDKN2A Antibody [NB200-106] - Analysis of FFPE mouse spleen using p19ARF/CDKN2A antibody at 10 ug/mL on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Nuclear staining in the germinal centers of spleen was observed. Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue).
