E-Cadherin Antibody (A0-G11-2-R)

Western Blot: E-Cadherin Antibody (A0-G11-2-R) [NBP3-32289] - Western blot analysis of E-Cadherin on different lysates with Mouse anti-E-Cadherin antibody (NBP3-32289) at 1/1,000 dilution. Lane 1: A431 cell lysate Lane 2: SW480 cell lysate Lane 3: MCF7 cell lysate Lysates/proteins at 10 ug/Lane. Predicted band size: 97.5 kDa Observed band size: 130 kDa Exposure time: 20 seconds; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32289) at 1/500 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Mouse IgG - HRP Secondary Antibody at 1:150,000 dilution was used for 1 hour at room temperature.

Immunohistochemistry: E-Cadherin Antibody (A0-G11-2-R) [NBP3-32289] - Immunohistochemical analysis of paraffin-embedded human breast carcinoma tissue with Mouse anti-E-Cadherin antibody (NBP3-32289) at 1,000 dilution. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 2 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32289) at 1/400 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

E-cadherin, or epithelial cadherin, is an cell-cell adhesion glycoprotein important to cellular processes such as morphology, polarity, development, tissue integrity and migration. E cadherin's extracellular domain interacts with other E-cadherin protein on adjacent epithelial cells to establish adhesion between them. E cadherin has also been identified as a tumor and metastasis suppressor, and its mutations have been linked to various forms of cancer.
