TRPM7 Antibody

Western Blot: TRPM7 Antibody [NBP1-46829] - Whole cell lysate (50 ug) from K-562, MCF-7, HeLa, and NIH 3T3 cells prepared using NETN lysis buffer. Antibody: Affinity purified rabbit anti-TRPM7 antibody used for WB at 1 ug/ml. Detection: hemiluminescence with an exposure time of 3 minutes.

Immunoprecipitation: TRPM7 Antibody [NBP1-46829] - Samples: Whole cell lysate (5, 15 and 50 ug for WB; 1 mg for IP, 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-TRPM7 antibody used for WB at 0.04 ug/ml (A) and 0.4 ug/ml (B) and used for IP at 3 ug/mg lysate. TRPM7 was also immunoprecipitated by rabbit anti-TRPM7 antibody BL9785, which recognizes an upstream epitope. Detection: Chemiluminescence with exposure times of 3 minutes (A) and 1 minute (B).

Immunoprecipitation: TRPM7 Antibody [NBP1-46829] - Detection of human TRPM7 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-TRPM7 antibody NBP1-46829 (lot NBP1-46829-3) used for IP at 6 ug per reaction. TRPM7 was also immunoprecipitated by a previous lot of this antibody (lot NBP1-46829-2). For blotting immunoprecipitated TRPM7, NBP1-46829 was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.

Background

TRPCs, mammalian homologs of the Drosophila transient receptor potential (trp) protein, are ion channels that are thought to mediate capacitative calcium entry into the cell. TRP-PLIK is a protein that is both an ion channel and a kinase. As a channel, it conducts calcium and monovalent cations to depolarize cells and increase intracellular calcium. As a kinase, it is capable of phosphorylating itself and other substrates. The kinase activity is necessary for channel function, as shown by its dependence on intracellular ATP and by the kinase mutants.[supplied by OMIM]
