TLR9 (Toll-like receptor 9) belongs to TLRs proteins which are pattern recognition receptors known to recognize a wide array of pathogen-associated molecular patterns (PAMPs) for the initiation of innate immune responses upon pathogen recognition. It is highly expressed in spleen, lymph node, tonsil and peripheral blood leukocytes, especially in plasmacytoid pre-dendritic cells. TLR9 is located intracellularly on the ER membrane, endosomes, lysosomes and phagosomes, however, has been reported to express extracellularly in lung tissue. It relocalizes from ER to endosome and lysosome upon stimulation with agonists. TLR9 is known to recognize microbial unmethylated 2'-deoxyribo cytidine-phosphate-guanosine DNA (CpG DNA). TLR9 acts via MYD88 and TRAF6, leading to NFkB activation, cytokine secretion followed by inflammatory response and has been shown to control lymphocyte response to Helicobacter infection. TLR9 interacts with MYD88, UNC93B1, CD300LH, BTK, CNPY3, HSP90B1 etc. and its interaction with CNPY3 /HSP90B1 is required for proper folding in ER and TLR9 interaction with CD300LH promote full activation of TLR9-triggered innate responses. The immune responses induced by TLR9 activation vary remarkably from one species to another e.g. stimulation is very potent in rodents, but less so in large animals including humans presumably due to differences in TLR9 expression between rodents and mammals.
