PUMA Antibody Summary

Immunogen
A portion of amino acids 186-199 (PLPRGHRAPEMEPN ) of human PUMA a was used as the immunogen.
Specificity
The amino acid sequence used as immunogen is 100% homologous in human (PUMA a and b) and 78% homologous in mouse and rat. Human PUMA a is a 193 amino acid protein with a predicted molecular weight of approximately 22 kD. Human PUMA b is a 131 amino acid protein with a predicted molecular weight of approximately 15 kD.

Background

A novel p53 inducible pro-apoptotic gene was identified recently and designated PUMA (for p53 upregulated modulator of apoptosis) in human and mouse. The PUMA gene encodes two BH3 domain-containing proteins termed PUMA-alpha and PUMA-beta. PUMA proteins bind Bcl-2, localize to the mitochondria, and induce cytochrome c release and apoptosis in response to p53. PUMA may be a direct mediator of p53-induced apoptosis.

Pathway Highlight: Which caspase substrates contribute to the morphological features associated with apoptosis?
Apoptosis, or programmed cell death, is controlled by a caspase signal cascade that activates downstream signals to induce the morphological changes used to differentiate apoptosis from other forms of cell death.  Novus Biologicals offers a variet...  Read full blog post.

Altered expression of BCL2 in cancer
Similar to other cell processes, the balance between cell survival and cell death is an important equilibrium that when altered expression of genes can lead to a variety of disease.  For example, too little cell death can promote cell overgrowth a...  Read full blog post.

Understanding Noxa Regulation of Apoptosis
Noxa is a pro-apoptotic gene belonging to the Bcl2 protein family that is unique in that it contains only BH3 domain. The BH3-only subclass of proteins, including proteins like PUMA and Bim in addition to Noxa, regulate the remaining Bcl-2 family memb...  Read full blog post.
