Application Notes
In Western blot this antibody detects a band at approximately 135 kDa. This is a high affinity, high titer antibody. Optimal dilutions should be determined by the end user. (Provided dilutions serve only as a suggestive starting point.).

E-Cadherin, a member of the cadherin superfamily, is implicated as a key player in different cellular processes including development, morphology, polarity, migration and tissue integrity (1-2). E-cadherin is a glycoprotein with an extracellular domain that interacts with other E-cadherin molecules on adjacent cells, thereby establishing adhesion between epithelial cells. Established as a tumor and metastasis suppressor, E-cadherin expression is frequently down-regulated or extinguished in malignancy which strongly correlates with poor prognosis (3-4). Mutations in E-cadherin are found in gastric, breast, colorectal, thyroid and ovarian cancer (5).

  1. Hello. I am currently working on Western Blots of serum with E-Cadherin N-terminus antibodies from other companies. Although a 55kDa splice fragment is not reported in the literature, I continue to obtain this band in my experimental blot, and not in my negative To work this out, I am looking for a monoclonal antibody specific to the N-terminus, and also an available blocking peptide to determine whether the band is specific or not. I am looking at your product EP700Y. It appears is it generated from a peptide immunogen. Is this peptide available for purchase?
    • Unfortunately we do not have this peptide available for purchase or to send out.
  2. I am using the E-cadherin antibody from your company (EP700Y) and the product information sheet indicates that the antibody detects a band at 135 kDa. But in my western I get a band around 100 kDa & other literature sources suggest the same. Can you please clarify this for me? Thank you.
    • E-cadherin is an 882 amino acids long protein with predicted molecular size of 97.4 kDa. However, this protein undergoes different post-translational modifications including glycosylation and the post-translationally modified protein may run at higher than expected molecular weight position. In our WB validation assays, most of our E-Cadherin antibodies including our best-seller #NBP2-19051, detects this target between 100-140 kDa positions. Therefore, we can assure you that your observed band at/above ~100 kDa is a specific band for the full length E-Cadherin protein. Please note that under some situations such as apoptosis or calcium influx, E-Cadherin protein undergoes molecular cleavage and its processing results in three C-terminal fragments (E-Cad/CTF1, E-Cad/CTF2, E-Cad/CTF3). These cleaved fragments may run below ~40 kDa position in the WB assay (PMID 16417575; PMID 13678586; PMID 11953314).
