Immunocytochemistry/Immunofluorescence: Angiopoietin-2 Antibody (117) [NBP2-89600] - Staining of Human Ang2 in A549 cells. Cells were fixed with 4% PFA, permeabilzed with 0.3% Triton X-100 in PBS, blocked with 10% serum, and incubated with NBP2-89600 antibody (1:300) at 4c overnight. Then cells were stained with the Alexa Fluor(R) 549-conjugated Goat Anti-rabbit IgG secondary antibody (red) and counterstained with DAPI (blue).

Background

Angiopoietin 1 and Angiopoietin 2 are important for development of the endothelium, by regulating tyrosine phosphorylation of the membrane receptor Tie 2/Tek. Angiopoietin 2 is only 60% homologous with Angiopoietin 1. Angiopoietin 1 binding to Tie 2/Tek causes phosphorylation of the receptor. Angiopoietin 2 competes for this binding, and thus blocks receptor phosphorylation. Angiopoietin 2 expression occurs at sites of vascular remodeling: dorsal aorta and major aortic branches, ovary, placenta and uterus.
