![Immunocytochemistry: N-Cadherin Antibody (13A9) [NBP1-48309] - Human meningioma mouse xenograft model KCI-MENG1-LPSX generated with the spontaneously immortal cell line KCI-MENG1-LP. Tumors from immunocompromised SCID mice were dissected and the derivative cell line KCI-MENG1-LPSX CL was generated. The EMA, PR, and N-cadherin IHC of the mouse tumor highly resembled the original patient-derived tumor. The vimentin- and Ki-67-stained cells in the mouse tumor tissue were markedly more abundant and more intensely stained than in the original tumor. KCI-MENG1-LPSX CL cells displayed the same patterns of immunostaining as the high passage parent cell line KCI-MENG1-HP, including the loss of PR staining. Scale bar 50 um. CAL27cis & Detroit‐562cis were cultured with IL‐6 (10 ng/mL, human recombinant IL‐6, R&D Systems, Minneapolis, USA) for 48 h (C). An anti‐human IL‐6 polyclonal antibody (R&D Systems, Minneapolis, USA) was used to neutralize the biological activities of IL‐6 (E, F). To inhibit signal transduction, CAL27cis & Detroit‐562cis cells were pre‐incubated for 1 h with STAT3 inhibitor V (Sigma‐Aldrich, Germany, 500 nmol/L), & then incubated with recombinant human IL‐6 (10 ng/mL) for 24 h before analysis. NIH/3T3 cells transiently expressing hPARM-1-GFP or deltaCT-GFP were fixed, immunostained for caveolin-1 (1:100, Novus Biologicals), and examined by confocal fluorescence microscopy. For hPARM-1-GFP-caveolin-1 co-localization, cells that demonstrated cell membrane PARM-1 localization were chosen. All co-localizations were observed following merging images of GFP-tagged proteins with those of Golgi, endosomes, plasma membrane, alpha-tubulin or caveolin-1 labeling.
