Western Blot: Sirtuin 1/SIRT1 Antibody [NBP2-27205] - Analysis of SIRT1 using SIRT1 antibody. HeLa lysate probed with 1) prebleed, 2) SIRT1 antibody and 3) mouse NIH 3T3 lysate probed with SIRT1 antibody at2 ug/ml. I goat anti-rabbit Ig HRP secondary antibody and PicoTect ECL substrate solution were used for this test.

Background

SIRT1 is a nuclear protein and member of the sirtuin protein family of deacetylating proteins. SIRT1 is involved in regulating a number of cellular pathways including apoptosis and cell senescence, longevity and aging (1-3). Not surprisingly, SIRT1 targets to forkhead (FOXO) transcription factors. SIRT1 physically binds to the p65/RelA protein in the NF-kB complex and inhibits the transcriptional activity by deacetylating the p65 component (4). SIRT1 anti-inflammatory responses have also been demonstrated using resveratrol, a potent activator of SIRT1, which has been shown to potentiate the SIRT1-mediated anti-inflammatory response (5). Detailed involvement of SIRT1 and NF-kB, and the control and interconnection of the Innate Immune and inflammatory response and aging, is reviewed by Salminen et al (6).
