Flow Cytometry: IL-9 Antibody (D9302C12) - Chimeric [NBP3-12021] - Mouse splenocytes were stimulated with PMA and Ionomycin in presence of Monensin for 4 hours. The cells were fixed using 2% PFA, permeabilized using 0.5% Triton and stained with anti-Fluorescein IgG antibody (4-4-20; isotype control, black line) or the rabbit IgG version of D9302C12 (NBP3-12021, blue line) at a dilution of 1:100 for 1h at RT. After washing, bound antibody was detected using a goat anti-mouse IgG AlexaFluor(R) 488 antibody at a dilution of 1:1000 and cells were analyzed using a FACSCanto flow-cytometer.

Background

IL-9 is a cytokine with immunoregulatory function, produced by activated T-helper type 2 lymphocytes. It acts on various cell types such as T- and B-lymphocytes, mast cells, haemopoietic progenitors and megakaryoblastic leukemia cell lines. IL-9 stimulates cell proliferation and prevents apoptosis. IL-9 may play a role in the pathogenesis of diseases such as certain lymphomas (Hodgin's disease, anaplastic lymphoma) and asthma. Although murine IL-9 can act on human cells, human IL-9 is inactive on mouse cells.
