Application Notes | This chimeric rabbit antibody was made using the variable domain sequences of the original Human IgG1 format, for improved compatibility with existing reagents, assays and techniques. The clinically used format of this antibody (which underwent additional de-fucosylation to increase antibody-dependent cellular cytotoxicity (ADCC)), has significant clinical activity in heavily pretreated patients with mycosis fungoides and Sezary syndrome, and is associated with an overall response rate of 36.8% and a median duration of response of 10.4 months (Duvic et al, 2014). It was shown to CCR4-expressing cells by antibody-dependent, cell-mediated cytotoxicity (ADCC). Therapeutically, this results in a dual mechanism of action; in addition to directly targeting malignant T cells over-expressing CCR4, this antibody also depletes T reg cells, an important therapeutic target in human cancers. This antibody is well-tolerated, and administration does not appear to trigger clinically significant autoimmune complications in phase 1/2 trials (Duvic et al, 2014). This antibody has been approved in Japan for the treatment of relapsed or refractory adult T-cell leukemia/lymphoma, and is in clinical trials for solid cancers. |
