![Immunohistochemistry: AIF-1/Iba1 Antibody [NB100-1028] - Temporal activation of caspase-8 in Iba1-positive myeloid cells. Frozen tissue sections from mice upon pMCAO (n = 9) were analyzed by immunohistochemistry for expression of active caspase-8 or for expression of the microglia/macrophage marker Iba1. The contralateral side to the ischemic area was used an internal control for each animal. A small increase in body size of Iba1 positive cells was detected in the peri-infarct area at 6 h, with larger increase in cell body & numbers of cells expressing Iba1 detected at 24 & 48 h post occlusion (a). Cleaved caspase-8 was detected at low levels at 6 h, but a significant increase in expression levels was observed at 24 & 48 h in the peri-infarct area, but not at the contralateral side (a). Double immunofluorescent staining revealed that increased active caspase-8 expression was localized in Iba1-positive cells (b). ![Immunohistochemistry: Goat Polyclonal AIF-1/Iba1 Antibody [NB100-1028] - Pax5 and Iba1 immunoreactivity in a formalin fixed paraffin embedded classic Hodgkin's lymphoma biopsy. Formalin fixed paraffin embedded sections were deparaffinized and rehydrated. Sections then underwent heat induced epitope retrieval in a citrate-based solution for 20min in a food steamer. Sections were cooled, then blocked in phosphate buffered saline containing 0.2% Triton-X100 and 10% normal donkey serum for 1h at room temperature. Sections were then incubated with the primary antibody for 16h at room temperature. The following day slides were washed three times with PBS containing 0.05% Tween-20. Sections were then incubated with donkey anti-goat (647 conjugate) at 5ug per mL for 45min at room temperature. Slides were then washed as above and mounted with a coverslip. ![Western Blot: PD-L1 Antibody - BSA Free [NBP1-76769] - Upregulated expression of PD‐L1 in cisplatin‐resistant cells through IL‐6/STAT3. The cisplatin resistant CAL27cis & Detroit‐562cis cells had increased IL‐6 mRNA & IL‐6 protein production compared to parental cells by qRT‐PCR & ELISA (A, B). CAL27cis & Detroit‐562cis were cultured with IL‐6 (10 ng/mL, human recombinant IL‐6, R&D Systems, Minneapolis, USA) for 48 h (C). An anti‐human IL‐6 polyclonal antibody (R&D Systems, Minneapolis, USA) was used to neutralize the biological activities of IL‐6 (E, F). To inhibit signal transduction, CAL27cis & Detroit‐562cis cells were pre‐incubated for 1 h with STAT3 inhibitor V (Sigma‐Aldrich, Germany, 500 nmol/L), & then incubated with recombinant human IL‐6 (10 ng/mL) for 24 h before analysis. ![Western Blot: PD-L1 Antibody - BSA Free [NBP1-76769] - LfcinB reduced IL‐6‐dependent PD‐L1 expression in cisplatin‐resistant HNSCC cells (A‐C) & inhibited the xenograft growth from CAL27cis & Detroit‐562cis (D, E). The markedly decreased IL‐6 (A) by ELISA & PD‐L1 by WB (B, C) in the acquired cisplatin‐resistant cells were detected. To investigate the effects of LfcinB on in vivo growth of CAL27cis & Detroit‐562cis, we treated nude mice carrying CAL27cis (D) & Detroit‐562cis (E) xenografts with once‐daily injection of 0.75 mg LfcinB (n = 5). Negative (LPS-, LPS-/CP7+, LPS-/CP7+) and positive (LPS+) controls were also included. Supernatants were harvested after 6 h and murine TNF-alpha measured by ELISA. After 16 h, cells were preincubated with various concentrations of inhibitor or control peptides (CP) for 1 h. Cells were then stimulated with 10 ng/ml LPS for 24 h. The secreted alkaline phosphatase (SEAP) was analyzed using the SEAPorter Assay Kit.
