Background

CD63 (LAMP-3, lysosome-associated membrane protein-3), a glycoprotein of tetraspanin family, is present in late endosomes, lysosomes and secretory vesicles of various cell types. It is also present in the plasma membrane, usually following cell activation. Hence, it has become an widely used basophil activation marker. In mast cells, however, CD63 exposition does not need their activation. CD63 interacts with integrins and affects phagocytosis and cell migration, it is also involved in H/K-ATPase trafficking regulation of ROMK1 channels. CD63 also serves as a T-cell costimulation molecule. Expression of CD63 can be used for predicting the prognosis in earlier stages of carcinomas.

CD63 antibody (clone H5C6) was used for FLOW application on human peripheral blood monocytes which were isolated from peripheral whole blood of healthy volunteers and the assay implicated - culture of adherent monocytes in the presence or absence of 10 ug/ml of Con A lectin, harvesting followed by washing with B/B/N buffer ( balanced salt solution/BSS with 0·2% BSA and 0·1% azide), incubation of 2 × 10^5 cells with 10ug of primary antibody or IgG1 isotype control on ice for 45 minutes, 2X washing with B/B/N buffer followed by 30 minutes incubation with FITC-labelled anti-mouse immunoglobulin, washing and analysis on flow cytometer (data not shown). The antibody was also used in functional/in-vitro assay for analysing the effects of anti-tetraspanin immunoglobulins on concanavalin A (Con A) induced monocyte fusion / multinucleated giant cells formation (Figure 2 and Figure 3), and for WB analysis of tetraspanin recombinant protein- extracellular domain-GST (Figure S1).

CD63 antibody (clone H5C6) was used for ICC-IF staining of primary NPCD/Niemann-Pick type C-1 deficient and normal fibroblasts that were infected with NL4.3-VSV-G-pseudotyped virus for 96 hours. The immunoassay involved - fixation of cells in 2% paraformaldehyde for 20 minutes, blocking with 5% normal goat serum containing 1% BSA, permeabilization with 0.1% saponin, use of primary antibody at 10 ug/ml concentration with 1 hour incubation, detection with Texas Red-conjugated goat anti-mouse secondary antibody (FIG 5).
