![<b>Biological Strategies Validation. </b>Western Blot: HIF-1 alpha Antibody [NB100-479] - Normoxic pre-treatment sensitizes glioma cells to radiation after graded chronic hypoxic (GCH) exposure. The results of anchorage-independent colony forming assays are shown for U87, U87-luc, GL261 glioma cells and 0308 GSCs after 5 Gy radiation exposure under varying O2 conditions. For easier comparisons among cell types, raw values are expressed as a % of the cell type's negative (non-irradiated) control and the means & SEMs are plotted. Each result represents at least 3 independent samples, plated in triplicate. Holm-Sidak comparisons for multiple groups were used for statistical comparisons of raw values (*p<0.05, **p<0.01). ![Immunocytochemistry/Immunofluorescence: HIF-1 alpha Antibody [NB100-479] - Immunofluorescence (IF) labeling and confocal microscopy analysis of the distribution of HIF-1alpha (A1-D1) and Abeta (A2-D2) in APPsw cells. DFO treatment significantly increased HIF-1alpha IF (D1) and reduced the Abeta IF (D2) in APPsw cells compared with controls (A1,A2). (A3-D3) DAPI was used to detect the nucleus (blue). (A4-D4) Merged images from the double channels indicated that DFO has an inhibitory effect on Abeta formation, and is involved in HIF-1alpha stabilization in APPsw cells. Fe treatment (100 uM) decreased HIF-1alpha (B1) and increased the Abeta levels (B2). Upon quantification, 100ug of protein was fractionated on 7% polyacralymide gel. Gel was transferred overnight onto nitrocellulose membrane. ![Flow Cytometry: TLR9 Antibody (26C593.2) [NBP2-24729] - Gating strategy for the detection of TLR-7 and -9 in B cell subsets. PBMC were isolated from whole blood and stained for surface markers before cells were fixed, permeabilised and stained for TLR-9. FSC and SSC were first used to gate out debris and SSC-A and SSC-H was utilized to eliminate duplicates. Further gating was done on CD45 and CD19, to target B cells. To separate between the different B cell populations, we gated on CD27 and IgD (D), followed by TLR-9 expression on these subsets. Data from one representative patient is shown. ![Immunocytochemistry/Immunofluorescence: IKK alpha Antibody (14A231) - BSA Free [NB100-56704] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton-X100. The cells were incubated with anti-IKK alpha Antibody (14A231) at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue).
