![Immunocytochemistry/Immunofluorescence: CD31/PECAM-1 Antibody [NBP1-71663] - HUVEC cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with CD31/PECAM-1 Antibody at 5 ug/mL overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). ![Immunohistochemistry: FIH-1/HIF-1AN Antibody [NB100-428] - 3D target protein expression in multicellular tumor spheroids3D expression analysis was carried out in the MCF-7, MDA-MB-231 & HBL-100 cell lines using multicellular tumor spheroids. The spheroids were cultured in spinner flasks for 1 week before fixation. Hypoxyprobe was used to examine the formation of hypoxic areas within the spheroids. NHE1, CAIX, MMP14 & FIH-1 expression was also analyzed. Incubation for an hour with PBS instead of primary antibody acted as a control. ![Immunocytochemistry/Immunofluorescence: Nestin Antibody (4D11) [NBP1-92717] - Mixed cultures of neonatal rat neurons and glia stained with NBP1-92717 (red), chicken antibody to Vimentin (NB300-223, green) and DNA (DAPI stain, blue). Astrocytes and neuronal stem cells stain strongly and specifically in a clearly filamentous fashion with the Nestin (4D11) antibody. The filamentous staining pattern is as expected as both Nestin and Vimentin are components of 10nm filaments. Note that some cells contain Nestin, but do not stain strongly for Vimentin and so appear red. Others contain Vimentin and not Nestin and so appear green - these are likely to be fibroblastic or endothelial cells. ![Gel Super Shift Assays: HIF-1 alpha Antibody [NB100-654] - Mouse HIF-1 alpha+/+ and HIF-1 alpha+/- cells were exposed to hypoxia or 125 uM CoCl2 for 6 h, nuclear extracts were then prepared and EMSA carried out. HIF-1 isolated from hypoxia or CoCl2-treated cells bound to the wild-type probe (lanes #1-6) but not to the mutant probe (lanes #7-12). HIF-1/DNA complex was detected in hypoxia (H)-or CoCl2 (Co)-treated cells (lanes #2, 3, 5, 6) but not in control (C) cells (lanes #1, 4). More complex (darker band) was seen in hypoxia-or CoCl2-treated HIF-1 alpha+/+ cells (lanes #2, 3) than that in hypoxia-or CoCl2-treated HIF-1 alpha+/- cells (lanes #5, 6). Supershift assay showed that the HIF-1/DNA complex was shifted up in the presence of wild-type (wt) oligo probe and 4 ug HIF-1 alpha antiboby (lanes #13 & 14).
