![Immunocytochemistry/Immunofluorescence: Nestin Antibody (4D11) [NBP1-92717] - Mixed cultures of neonatal rat neurons and glia stained with NBP1-92717 (red), chicken antibody to Vimentin (NB300-223, green) and DNA (DAPI stain, blue). Astrocytes and neuronal stem cells stain strongly and specifically in a clearly filamentous fashion with the Nestin (4D11) antibody. The filamentous staining pattern is as expected as both Nestin and Vimentin are components of 10nm filaments. Note that some cells contain Nestin, but do not stain strongly for Vimentin and so appear red. Others contain Vimentin and not Nestin and so appear green - these are likely to be fibroblastic or endothelial cells. ![Gel Super Shift Assays: HIF-1 alpha Antibody [NB100-654] - Mouse HIF-1 alpha+/+ and HIF-1 alpha+/- cells were exposed to hypoxia or 125 uM CoCl2 for 6 h, nuclear extracts were then prepared and EMSA carried out. HIF-1 isolated from hypoxia or CoCl2-treated cells bound to the wild-type probe (lanes #1-6) but not to the mutant probe (lanes #7-12). HIF-1/DNA complex was detected in hypoxia (H)-or CoCl2 (Co)-treated cells (lanes #2, 3, 5, 6) but not in control (C) cells (lanes #1, 4). ![Immunohistochemistry-Paraffin: AKT1 [p Ser473] Antibody (104A282) [NB100-56749] - IHC analysis of an FFPE human breast carcinoma tissue section using 1:250 dilution of pSer473 AKT1 antibody (clone 104A282) on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) with 10 mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching using peroxide block. The sections were incubated with primary antibody for 30 minutes. Bond Polymer Refine Detection (Leica Biosystems) and DAB were used for signal detection which followed counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) were performed using Aperio AT2 (Leica Biosystems). This antibody generated a diffused cytoplasmic staining of phosphor-AKT (Ser-473) in the cancer cells as well as the stromal cells. Some cancer cells depicted nuclear stianing also.
