The TLR10 reporter line may be useful for TLR10-based functional studies. The functional activity of the NF-kB/SEAP reporter in the TLR10 stable cell line has been established and validated through PMA activation of the NF-kB pathway (Figure 1). Hence, the cell line may be useful for reseasrchers screening for putative TLR10 ligands activating the NF-kB pathway.
Any specific ligand for TLR10 is still unknown. However, TLR10 can indeed activate the NF-kB signaling pathway through direct association with the common Toll/IL-1 receptor domain adapter, MyD88, according to a recent study utilizing a chimeric CD4/TLR10 molecule. TLR10 expression in the TLR10/NF-kB/SEAPorter HEK 293 cell line has been validated by Western blotting (Fig. 2) and flow cytometry (Fig. 3).

Notes

- Wash hands after handling the TLR10 reporter line and before leaving the lab.

Background
Toll-like receptors (TLRs) are signaling molecules that recognize different microbial products during infection and serve as an important link between the innate and adaptive immune responses. These proteins act through adaptor molecules such as MyD88 and TIRAP to activate various kinases and transcription factors. TLR10 is highly homologous to TLRs 1 and 6 and is most highly expressed in lymphoid tissues. Most recently genetic variation in TLR10 has been associated with contributing to asthma risk. It should be noted that TLR10 exists in mice only as a pseudogene.
