Background
Thanatos-associated protein (THAP) domain is an evolutionarily conserved C2-CH (C-X2-4-C-X35-50-C-X2-H) zinc finger motif which has sequence-specific DNA-binding activity and twelve THAP proteins, each containing an N-terminally located THAP domain, have been identified in humans (THAP0 to THAP11), some of which (THAP0, -1, -2, -4, -7, and -11) are also conserved in mice/rats. Individual THAP proteins have been implicated in a diverse array of physiological processes, including cell proliferation, regulation of transcription, apoptosis, and maintenance of embryonic stem (ES) cell pluripotency. THAP11 is regulated by shuttling between cytoplasm - nucleus, and is considered a transcriptional repressor that plays a central role in embryogenesis as well as in ES cell pluripotency. It act as a sequence-specific DNA-binding factor which represses gene expression in pluripotent ES cells by directly binding to key genetic loci and recruiting epigenetic modifiers. THAP11 has been suggested as a candidate for polyglutamine disorders based on polymorphism and protein-folding simulation studies. RONIN is the mouse homolog of THAP11 and its homozygous deletion happened to be embryonically lethal in mice wherein was shown to be essential factor underlying embryogenesis/ES cell pluripotency via its activational/ repressional effects on transcription. THAP11 also functions as a negative regulator of cell growth in human HepG2 hepatoma cells through transcriptional repression of the proto-oncogene MYC.
