Background

This gene encodes a pyrin-like protein containing a pyrin domain, a nucleotide-binding site (NBS) domain, and a leucine-rich repeat (LRR) motif. This protein interacts with the apoptosis-associated speck-like protein PYCARD/ASC, which contains a caspase recruitment domain, and is a member of the NALP3 inflammasome complex. This complex functions as an upstream activator of NF-kappaB signaling, and it plays a role in the regulation of inflammation, the immune response, and apoptosis. Mutations in this gene are associated with familial cold autoinflammatory syndrome (FCAS), Muckle-Wells syndrome (MWS), chronic infantile neurological cutaneous and articular (CINCA) syndrome, and neonatal-onset multisystem inflammatory disease (NOMID). Multiple alternatively spliced transcript variants encoding distinct isoforms have been identified for this gene. Alternative 5' UTR structures are suggested by available data; however, insufficient evidence is available to determine if all of the represented 5' UTR splice patterns are biologically valid. FAQs for NLRP3/NALP3 Recombinant Protein (H00114548-P01). (Showing 1 - 5 of 5 FAQs).

  1. Are the NLRP3/NALP3 antibodies validated in Simple Western?
    • Yes, we offer a NLRP3/NALP3 antibody that have been tested in Simple Western: NBP2-03948.
  2. Does NLRP3/NALP3 antibodies come in lyophilized form?
    • Yes, we carry 3 NLRP2 antibodies in lyophilized format: AF6789, AF7010, MAB6789, MAB7578.
  3. What is the theoretical molecular weight for NLRP3/NALP3 antibodies?
    • The TMW of NLRP3/NALP3 antibodies is approximately 118 kDa.
  4. I want to use NLRP3 to stimulate a human cell line to check whether there is expression of IL-1beta or IL-18. Should I choose the partial or full-length NLRP3 protein? Do you have recommended dosage of NLRP3 for cell culture? Thanks.
    • This product is produced by a Taiwanese company called Abnova and we distribute it for them. They do not test the activity of their recombinant proteins. The proteins are produced in a cell-free wheat germ system with an N-terminal 26kDa GST-tag. Due to this expression system, in our experience, the proteins do not always undergo the same folding and post-transnational modifications they might undergo in an endogenous cell. Because of this, we do not recommend this protein for use in functional assays or cell culture and do not guarantee them for this use. They are mainly intended for use in Western Blots as a positive control with their antibody.
  5. I'm using the Nalp3 recombinant protein as a positive control in a Western Blot. How much protein do you suggest I load onto the gel?
    • Abnova, the manufacturer of H00114548-P01, suggests loading 0.1-0.2ug per lane of purified protein in Western blot.
