Source and Preparation: MMP2 (2C1) hybridoma culture medium. The hybridoma was grown in medium supplemented with Ig-depleted fetal bovine serum (serum passed through a Protein G column). The monoclonal mouse antibody was purified by Protein G affinity chromatography (and contains no detectable bovine immunoglobulins).

MMP2 (matrix metalloproteinase-2; CLG4A) is a ubiquitinous metalloproteinase enzyme belonging to peptidase M10A family which is implicated in versatile activities including vasculature remodeling, angiogenesis, tissue repair, tumor invasion, inflammation, and atherosclerotic plaque rupture. Besides degrading proteins in the extracellular matrix, MMP2 also acts on various nonmatrix proteins such as big endothelial 1, beta-type CGRP promoting vasoconstriction, KISS etc. MMP2 plays a key role in myocardial cell death pathways and also contributes to myocardial oxidative stress through regulation of GSK3-beta activity. PKC facilitates MDM2 phosphorylation which is crucial to its enzymatic activity. MMP2 propeptide is processed by MMP14/MT-MMP1 and MMP16/MT-MMP3, and a C-terminal autocatalytic cleavage generates PEX chain which exerts anti-angiogenic as well as anti-cancer properties alongwith inhibiting cell migration or cell adhesion to FGF2/vitronectin. MMP2's isoform 2 facilitates the proteolysis of CHUK/IKKA and initiates a primary innate immune response by inducing mitochondrial-nuclear stress signaling with activation of pro-inflammatory NFkB, NFAT and IRF transcriptional pathways. MMP2 is implicated in several pathologies including metastasis of cancer and defects in MMP2 have been linked to Torg-Winchester syndrome (TWS) which is also called as multicentric osteolysis nodulosis and arthropathy (MONA).

  1. Could you guarantee NB200-113 and NB200-114 both detect human pro-form and active form MMP2 ?
    • Yes, both of your mentioned antibodies (i.e. NB200-113 and NB200-114) detects the pro (approximately 70kD) as well as the activated forms (approximately 62kD) of MMP-2 in Western Blot. In fact, the validation image on the datasheet of #NB200-114 shows a very nice blot where these two forms are very evident and this antibody has been cited in 8 peer reviewed publications.
