Western Blot: Galectin-9 Antibody (PSH02-44) [NBP3-32531] - Western blot analysis of Galectin-9 on different lysates with Rabbit anti-Galectin-9 antibody (NBP3-32531) at 1/1,000 dilution. Lane 1: THP-1 cell lysate Lane 2: HL-60 cell lysate Lane 3: RAW264.7 cell lysate Lysates/proteins at 20 ug/Lane. Predicted band size: 92 kDa Observed band size: 92 kDa Exposure time: 1 minutes 59 seconds; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32531) at 1/1,000 dilution was used in 5% NFDM/TBST at 4 overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Flow Cytometry: Galectin-9 Antibody (PSH02-44) [NBP3-32531] - Flow cytometric analysis of THP-1 cells labeling Galectin-9. Cells were fixed and permeabilized. Then stained with the primary antibody (NBP3-32531, 1/1,000) (red) compared with Rabbit IgG Isotype Control (green). After incubation of the primary antibody at +4℃ for an hour, the cells were stained with a iFluor™ 488 conjugate-Goat anti-Rabbit IgG Secondary antibody at 1/1,000 dilution for 30 minutes at +4℃. Unlabelled sample was used as a control (cells without incubation with primary antibody; black).

Background

The galectins are a family of beta-galactoside-binding proteins implicated in modulating cell-cell and cell-matrix interactions. Galectin 9 is an S-type lectin and an eosinophil chemoattractant (ECA) produced by antigen-activated T cells. Galectin 9 is involved in apoptosis, is thought to be a prognostic factor for breast cancer and is also overexpressed in Hodgkin's disease tissue. The protein has N- and C- terminal carbohydrate-binding domains connected by a link peptide. Multiple alternatively spliced transcript variants have been found for this gene.
