Flow Cytometry: CD45RC Antibody (OX-22) - Chimeric - BSA Free [NBP3-43542] - Staining of rat (Rattus norvegicus) lymphocytes with either an isotype control (A) or NBP3-43542 (B) at a concentration of 1 ug/ml for 30 mins at RT. After washing, bound antibody was detected using an Alexa Fluor 488 conjugated donkey anti-rabbit antibody and cells analysed on a FACSCanto flow-cytometer.

• Specificity: OX-22 binds specifically to the high molecular weight form of the leucocyte common antigen, CD45RC. OX-22 and OX-31 compete for binding but do not compete with OX-32. The antigen is expresssed on the surface of B cells, around half of bone marrow cells, all CD8+ve T cells, and is able to differentiate between CD4+CD45RChigh (Th1-like) and CD4+CD45RClow (Th2-like) cell types. CD45 extracellular domain is able to interact with a number of different proteins including CD4, TCR, CD2, Thy-1 and CD26. CD45 is important for signalling throguh the T cell receptor and in the selection of T and B lymphocytes.

Background

The CD45 family of cell surface glycoprotein antigens exist as a set of isoforms. The patterns of monoclonal antibody recognition of the CD45 phenotypes can be restricted (CD45RA, RB and RC), or broader, such as the CD45 T 200 species. The "restricted" subsets of this group arise as a result of translation of alternatively spliced mRNAs from exons A, B and C (exons 4, 5 and 6), where exons 3-15 encode the extracellular domain of the CD45 molecule. The restricted epitopes are differentially expressed on T and B lymphocytes, including functionally distinct T cells. CD45 (LCA) is a transmembrane phosphotyrosine phosphatase expressed on leucocytes. Myeloid cells do not express the RC isoform.
