• Specificity: This monoclonal antibody recognizes a protein of 145kDa, identified as CD117/p145kit. It is found on a wide variety of tumor cells including follicular and papillary carcinoma of thyroid, adenocarcinomas from endometrium, lung, ovary, pancreas, and breast as well as malignant melanoma, endodermal sinus tumor, and small cell carcinoma. However, anti-CD117 has been particularly useful in differentiating gastrointestinal stromal tumors from Kaposi's sarcoma, tumors of smooth muscle origin, fibromatosis, and neural tumors of the GI tract. Anti-CD117 is also useful in recognizing myeloblasts in bone marrow biopsy and clot section.

• Application Notes: Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 1mM EDTA buffer, pH 9.5, for 45 min at 95C followed by cooling at RT for 20 minutes.
  Optimal dilution for a specific application should be determined.

Background

CD117 is also know as c-Kit. Antibodies to CD117 / c-Kit may be useful for the identification of several cancers including mast cell diseases, acute myeloid leukemia (AML), small cell lung carcinoma (SCLC), and Ewing's sarcoma, and in the differentiation between gastrointestinal stromal tumors (GISTs) and other intra-abdominal mesenchymal tumors
