BCR Antibody

Western Blot: BCR Antibody [NBP1-19098] - Whole cell lysate from HeLa and 293T cells. BCR was also immunoprecipitated by rabbit anti-BCR antibody NBP1-19097.

Immunocytochemistry/Immunofluorescence: BCR Antibody [NBP1-19098] - Human K562 cells (contains the chromosomal translocation, t(9:22) that creates the BCR/ABL fusion gene). Antibody used at a dilution of 1:1,000 (1ug/ml). Detection: Green-fluorescent goat anti-rabbit IgG - Alexa Fluor 488 used at a dilution of 1:1,000.

Flow Cytometry: BCR Antibody [NBP1-19098] - Flow Cytometric Detection of BCR. 1 x 106 K562 cells were fixed, permeabilized, and stained with 0.25 mcg anti-BCR in a 150 mcl reaction. Isotype anti-KLH control (red), no antibody (blue) and anti-BCR (black).

Background

A reciprocal translocation between chromosomes 22 and 9 produces the Philadelphia chromosome, which is often found in patients with chronic myelogenous leukemia. The chromosome 22 breakpoint for this translocation is located within the BCR gene. The translocation produces a fusion protein which is encoded by sequence from both BCR and ABL, the gene at the chromosome 9 breakpoint. Although the BCR-ABL fusion protein has been extensively studied, the function of the normal BCR gene product is not clear. The protein has serine/threonine kinase activity and is a GTPase-activating protein for p21rac. Two transcript variants encoding different isoforms have been found for this gene.
