Immunocytochemistry/Immunofluorescence: B220/CD45R Antibody (RA3-6B2) - Chimeric [NBP3-11980] - Immunofluorescence analysis of paraformaldehyde fixed mouse splenocytes immobilized on Shi-fix(TM) cover-slips and stained with the chimeric rabbit IgG version of RA3-6B2 (NBP3-11980) at 10 ug/ml followed by Alexa Fluor(R) 488 secondary antibody (2 ug/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-11980, DAPI, merged channels and an isotype control. The isotype control was stained with anti-Fluorescein antibody (NBP2-52638) followed by Alexa Fluor(R) 488 secondary antibody.

Flow Cytometry: B220/CD45R Antibody (RA3-6B2) - Chimeric [NBP3-11980] - Mouse splenocytes were stained with anti-Fluorescein IgG antibody (4-4-20; isotype control, black line) or the rabbit IgG1 version of RA3-6B2 (NBP3-11980, blue line) at a dilution of 1:100 for 1h at RT. After washing, bound antibody was detected using a goat anti-rabbit IgG AlexaFluor(R) 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Background

CD45 is a family of single chain transmembraneous glycoproteins consisting of at least four isoforms (220, 205, 190, 180 kDa) which share a common large intracellular domain. Their extracellular domains are heavily glycosylated. The different isoforms are produced by alternative messenger RNA splicing of three exons of a single gene on chromosome 1. CD45 is expressed on cells of the human hematopoietic lineage (including hematopoietic stem cells) with the exception of mature red cells. It is not detected on differentiated cells of other tissues. It is likely that CD45 plays an important role in signal transduction, inhibition or upregulation of various immunological functions. Antibodies recognizing a common epitope on all of the isoforms are termed CD45 whilst those recognizing only individual isoforms are termed CD45RA or CD45RO etc.
