Notes

HEK293T cells in 10-cm dishes were transiently transfected with a non-lipid polymer transfection reagent specially designed and manufactured for large volume DNA transfection. Transfected cells were cultured for 48hrs before collection. The cells were lysed in modified RIPA buffer (25mM Tris-HCl pH7.6, 150mM NaCl, 1% NP-40, 1mM EDTA, 1xProteinase inhibitor cocktail mix, 1mM PMSF and 1mM Na3VO4, and then centrifuged to clarify the lysate. Protein concentration was measured by BCA protein assay kit.This product is manufactured by and sold under license from OriGene Technologies and its use is limited solely for research purposes.

Background

Microtubules of the eukaryotic cytoskeleton perform essential and diverse functions and are composed of a heterodimer of alpha and beta tubulins. The genes encoding these microtubule constituents belong to the tubulin superfamily, which is composed of six distinct families. Genes from the alpha, beta and gamma tubulin families are found in all eukaryotes. The alpha and beta tubulins represent the major components of microtubules, while gamma tubulin plays a critical role in the nucleation of microtubule assembly. There are multiple alpha and beta tubulin genes, which are highly conserved among species. This gene encodes alpha tubulin and is highly similar to mouse and rat Tuba1 gene. Northern blotting studies have shown that the gene expression is predominantly found in morphologically differentiated neurologic cells. This gene is one of three alpha-tubulin genes in a cluster on chromosome 12q. [provided by RefSeq].   1. Our customer want to know which tubulin (alpha or beta) is suitable for WB as the loading control in brain tissue. Could you recommend the tubulin alpha or beta as a loading control for WB in brain tissue? In case of adipose tissue, tubulin alpha is not suitable as a loading control. How about in brain tissue? Could you let me know?
    • Both beta and alpha tubulin are expressed in the brain, and either one would make a suitable loading control. Beta tubulin may give a cleaner signal than alpha, but this varies greatly. You may wish to consider NB600-936.
  2. We are working with mouse liver tissue, will this alpha tubulin antibody work as a loading control?
    • Yes, alpha tubulin is expressed in nearly all tissue types including liver; however we do not recommend using as a loading control antibody in stomach or oral mucosa tissue as expression of alpha tubulin in these areas is minimal.
  3. Will your alpha tubulin antibody work as a loading control for simple western?
    • Yes, this alpha tubulin antibody has been validated in simple western and makes an excelent loading control antibody. Simple western has a lower detection limit of 12kDA and alpha tubulin detects around 55kDA.
  4. Will this alpha tubulin antibody recognize both isoforms of alpha tubulin?
    • The epitope for this alpha tubulin antiboyd lies on the C-terminus of the protein and the difference between the two major isoforms is witin the first 35 aa of the N-terminus so this alpha tubulin antibody will recognize both isoforms.
  5. We're looking at LC3B, aSYN and p62 which are 18, 23 and 65 kDA respectively so we need the loading control antibody to be between those.
    • Alpha tubulin antibody detects around 55kDA so that would be a good option for a loading control antibody for those targets as you should be able to easily distinguish from the other bands.
  6. I am looking for a primary alpha tubulin  antibody that is conjugated to a red fluorophore.
    • Okay, our most popular alpha tubulin antibody is NB100-690 and is available in a variety of formats. So if you are looking for red I would recommend a DyLight 550, 650 etc.
