CXCR4 protein is stained red, & MYH11 is stained green to indicate the PASMCs (×400; scale bars indicate 50 μm). CXCR4 fluorescence intensity was calculated (b; n = 3). CXCR4 protein expression levels in lung tissue were examined by western blot. GAPDH served as an internal control (c, d; n = 3). Data are presented as the mean ± SD. #Value significantly greater than the corresponding value in saline-treated normoxic mice (##P 50 CTCs/ 2.5 ml blood) was processed via ficoll density centrifugation & the PBMC fraction was collected. Immunomagnetic anti-CD45 depletion was performed on the obtained PBMCs & the remaining cells were washed, cytospunned onto the slides. Slides were stained for PSMA, EpCAM, sLex, & CXCR4 using the protocol as described in Figure S1. MDA, PC3, & KG1 cells were simultaneously stained as a control for the following markers: PSMA= Magenta, EpCAM= Yellow, HECA-452= Green, CXCR4= Red. ![<b>Biological Strategies Validation. </b>Immunohistochemistry-Paraffin: CCL2/MCP1 Antibody [NBP1-07035] - IHC analysis of a formalin-fixed paraffin-embedded (FFPE) human breast carcinoma tissue section using 1:1000 dilution of CCL2/MCP1 antibody (NBP1-07035) on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) with 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching using peroxide block. The sections were incubated with primary antibody for 30 minutes. Bond Polymer Refine Detection (Leica Biosystems) and DAB were used for signal detection which followed counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) were performed using Aperio AT2 (Leica Biosystems). This antibody generated a diffused cytoplasmic staining of CCL2 antigen in the cancer cells, stromal cells as well as the endothelial cells. The stroma itself showed a weak immunopositivity for CCL2. ![Immunocytochemistry/Immunofluorescence: RIPK3/RIP3 Antibody [NBP2-24588] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with RIPK3/RIP3 Antibody [NBP2-24588] at a 1:200 dilution overnight at 4C and detected with DyLight 488 (Green) at a 1:500 dilution. Alpha tubulin was used at a 1:1000 dilution as a co-stain and detected with DyLight 550 (Red) at a 1:500 dilution. Nuclei were detected with DAPI (Blue) at 2.0ug/ml in 1X PBS.
