Western Blot: Sortilin Antibody (JE60-00) [NBP3-32968] - Western blot analysis of Sortilin on different lysates with Rabbit anti-Sortilin antibody (NBP3-32968) at 1/1,000 dilution. Lane 1: SK-MEL-28 cell lysate (20 ug/Lane) Lane 2: Human lung tissue lysate (40 ug/Lane) Lane 3: Human kidney tissue lysate (40 ug/Lane) Lane 4: Human liver tissue lysate (40 ug/Lane) Lane 5: SH-SY5Y cell lysate (20 ug/Lane) Lane 6: Human brain tissue lysate (40 ug/Lane) Predicted band size: 92 kDa Observed band size: 100 kDa Exposure time: Lane 1-4: 1 minute 7 seconds; Lane 5-6: 2 minutes 37 seconds; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32968) at 1/1,000 dilution was used in 5% NFDM/TBST at 4C overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Immunohistochemistry: Sortilin Antibody (JE60-00) [NBP3-32968] - Immunohistochemical analysis of paraffin-embedded human brain tissue with Rabbit anti-Sortilin antibody (NBP3-32968) at 1/500 dilution. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32968) at 1/500 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

Functions as a sorting receptor in the Golgi compartment and as a clearance receptor on the cell surface. Required for protein transport from the Golgi apparatus to the lysosomes by a pathway that is independent of the mannose-6-phosphate receptor (M6PR). Also required for protein transport from the Golgi apparatus to the endosomes. Promotes neuronal apoptosis by mediating endocytosis of the proapoptotic precursor forms of BDNF (proBDNF) and NGFB (proNGFB). Also acts as a receptor for neurotensin. May promote mineralization of the extracellular matrix during osteogenic differentiation by scavenging extracellular LPL. Probably required in adipocytes for the formation of specialized storage vesicles containing the glucose transporter SLC2A4/GLUT4 (GLUT4 storage vesicles, or GSVs). These vesicles provide a stable pool of SLC2A4 and confer increased responsiveness to insulin. May also mediate transport from the endoplasmic reticulum to the Golgi.
