RUNX2 is a transcription factor that mediates the expression several downstream proteins, which affect osteoblast differentiation and mitotic progression. It also is important in monocyte activation, as it is upregulated in response to LPS infection. Through interaction with Twist 1 and Twist 2, RUNX2 regulates osteocalcin transcription, following parathyroid hormone signaling for bone loss repair. Ossification in skeletal formation is also partially regulated through RUNX2. Essentially, this transcription factor is responsible for many developmental and repair mechanisms in bone and collagen.
