Simple Western lane view shows lysates of BG01V human embryonic stem cells, loaded at 0.2 mg/mL. A specific band was detected for SOX2 at approximately 50 kDa (as indicated) using 4 µg/mL of Mouse Anti-Human/Mouse/Rat SOX2 Monoclonal Antibody (Catalog # MAB2018). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.
Western blot shows lysates of NTera‑2 human testicular embryonic carcinoma cell line, F9 mouse teratocarcinoma stem cells, D3 mouse embryonic stem cell line, and rat cortical stem cells. PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human/Mouse/Rat SOX2 Monoclonal Antibody (Catalog # MAB2018) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for SOX2 at approximately 36 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
E‑Cadherin and SOX2 were detected in BG01V human embryonic stem cells using 10 µg/mL Goat Anti-Human E‑Cadherin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF648) and 10 µg/mL Mouse Anti-Human/Mouse/Rat SOX2 Monoclonal Antibody (Catalog # MAB2018). Cells were incubated with primary antibodies for 3 hours at room temperature. Cells were stained for E‑Cadherin using the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; Catalog # NL001) and for SOX2 using the NorthernLights 493-conjugated Anti-Mouse Secondary Antibody (green; Catalog # NL009). The staining was developed using HRP labeled anti-mouse secondary antibody and DAB reagent, and nuclei of cells were counter-stained with hematoxylin. General cytoplasmic staining appears enriched in melanocytes below the epidermis. Some nuclear reactivity is observed as well.
Immunocytochemistry/Immunofluorescence: MITF Antibody (21D1418) [NB100-56561] - HeLa cells were fixed in 4% paraformaldehyde for 10 min and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-MITF Antibody (21D1418) NB100-56561 at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:500 dilution for 60 minutes. Cells were imaged using a 100X objective and digitally deconvolved.

Mouse splenocytes were fixed using formaldehyde, resuspended in lysis buffer, and sonicated to shear chromatin. HNF‑3 beta /FoxA2/DNA complexes were immunoprecipitated using 5 μg Goat Anti-Human HNF‑3 beta /FoxA2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2400) or control antibody (Catalog # AB-108-C) for 15 minutes in an ultrasonic bath, followed by Biotinylated Anti-Goat IgG Secondary Antibody (Catalog # BAF109). Immunocomplexes were captured using 50 μL of MagCellect Streptavidin Ferrofluid (Catalog # MAG999) and DNA was purified using chelating resin solution. The E-RABP promoter was detected by standard PCR.
Western blot shows lysates of HepG2 human hepatocellular carcinoma cell line, Jurkat human acute T cell leukemia cell line, and K562 human chronic myelogenous leukemia cell line.
