A recombinant protein corresponding to the Pyrin domain (PYD) of human NALP3 (amino acids 6-89) was used as the immunogen for this antibody. NALP3 belongs to a class of protein motifs known as the death fold. As a class, death fold motifs primarily function in regulating inflammatory and apoptosis responses. The PYD of NALP3 is required for association with ACS during inflammasome formation.

NLRP3 belongs to the NLR [nucleotide binding oligomerization domain (NOD) and leucine rich region repeat (LRR) containing proteins] family of innate immune cytoplasmic receptors (reviewed in De Nardo and Latz, 2011; Kanneganti, 2010; Anand, 2011). NLR family members contain an N-terminus effector region consisting of either a pyrin (PYD), a caspase activation and recruitment (CARD), or a baculorvirus inhibitor of apoptosis protein repeat (BIR) domain. NLRs also have an intermediate conserved NOD domain and a C-terminus ligand binding LRR domain. NLRs, like TLRs (Toll-like receptors), alert the mammalian immune system to the presence of PAMPs (pathogen associated molecular patterns) and DAMPs (danger associated molecular patterns). Like a number of other NLRs, NLRP3 functions in a multiprotein complex called the inflammasome. The inclusion of a particular NLR within an inflammasome depends upon the upstream trigger. The NLRP3 inflammasome is considered to be a global sensor and assembles in response to a broad spectrum of PAMPs and DAMPs. The PYD of activated NLRP3 associates with the PYD of ASC, triggering the CARD of ASC to associate with the CARD of caspase-1 and completes the assembly of the NLRP3 inflammasome. The inflammasome serves as a platform for caspase-1 activation. Active caspase-1 cleaves the proforms of the proinflammatory cytokines IL-1beta, IL-18, and IL-33 to their active, secreted forms. The mechanisms from NLRP3 activation to inflammasome formation and caspase-1 activation remain to fully elucidated.
