Background

Lysosomal associated membrane protein 2 (LAMP2), also known as CD107b and LGP110, is an approximately 110 kDa transmembrane glycoprotein that is a major component of lysosomal membranes (1). Mature human LAMP2 consists of a 347 amino acid (aa) intralumenal domain, a 24 aa transmembrane segment, and a 35 aa cytoplasmic tail (2). Its lumenal domain is organized into two heavily N-glycosylated regions separated by a Ser/Pro-rich linker that carries a minor amount of O‑linked glycosylation (2, 3). Alternate splicing generates a human LAMP2 isoform (LAMP2B) with a substituted juxtamembrane lumenal region, transmembrane segment, and cytoplasmic tail (4). Within the lumenal domain, human LAMP2 shares approximately 64% aa sequence identity with mouse and rat LAMP2. LAMP2 itself is subject to lysosomal degradation following cleavage of its lumenal domain (5). It mediates the lysosomal uptake of the chaperone HSC73 in complex with cargo proteins and is required for the lysosomal destruction of autophagic vacuoles (6, 7). In cytotoxic T cells and mast cells, LAMP2 is expressed in the membranes of intracellular granules that contain effector molecules such as perforin, granzymes, eicosanoids, and histamine (8-10). Up‑regulated LAMP2 at the plasma membrane serves as an indicator of cell activation of CD8+ T cells, mast cells, monocytes, and platelets (9-12). LAMP2 is a native ligand for lectins Galectin-1 and Galectin-3 (13‑15).

Western Blot: LC3A Antibody [NB100-2331] - High autophagosome concentration is consumed during early immortalized human mesenchymal stem cell differentiation. Immortalized human mesenchymal stem cells were differentiated under osteogenic conditions (see Materials and methods) and assayed for changes in LC3I and LC3II during a 72-hour window. Cells were differentiated under standard conditions (top) or with addition of 5 uM rapamycin (middle) or 5 nM bafilomycin (bottom) for the first 3 hours of differentiation to modulate autophagy. Immunoblots were performed for LC3 at the indicated time points to assess autophagosome degradation via relative changes in LC3II (lower band; 17 kDa). Studies were repeated three times with similar trends seen consistently. Western Blot: Proteinase 3/Myeloblastin/PRTN3 Antibody [NBP1-25966] - Human PMN (peripheral blood mononuclear cells isolated from buffycoat; denatured, reduced) using Rabbit antibody to c-terminal region of Pr3 (Wegener autoantigen): whole serum at 1: 500 dilution; blocked with 1% LFDM for 15 minutes at room temperature with shake, primary antibody incubated for 15 minutes at room temperature, washed 3 times with PBST, 5 minutes each. Secondary antibody was also incubated for 15 minutes at room temperature.
Immunocytochemistry/Immunofluorescence: Proteinase 3/Myeloblastin/PRTN3 Antibody [NBP1-25966] - Human PBMC were isolated and adjusted to 106 cells. Cells were fixed with 2% formaldehyde for 10 min at 37C. Washed twice with PBS before cytospin the cells onto microscope slides. Cells were blocked with PBS containing 1%BSA for 20 min at RT. Excess of blocking solution was removed and cells were then incubated with Rabbit Ab to c-terminal region of Pr3 (Wegener autoantigen): whole serum for 30 min at RT (diluted 1:100 in the blocking buffer). Washed 3X with PBS and incubated with anti-Rabbit Alexa 586 for further 30 min. Washed as before and nuclear counterstained with DAPI. Neutrophils and Monocytes, known to have PR3 are intensely stained by the Rabbit Ab to c-terminal region of Pr3 (Wegener autoantigen): whole serum.
