This IKEPP antibody is useful for Western blot, where a single band of ~60 kDa is detected. It is not useful for Immunocytochemistry/Immunofluorescence. The Na+/H+ exchange regulatory factor (NHERF) family of PDZ-containing proteins comprises of four members including NHERF1/EBP50, NHERF2/E3KARP, NHERF3/PDZK1 and NHERF4/IKEPP/PDZK2; and among all, NHERF1/EBP50 and NHERF2 are most widely distributed. NHERF1/EBP50 and NHERF2 contain only two PDZ domains each, whereas, the highly homologous PDZK1 and IKEPP each contain four PDZ domains. IKEPP (intestinal and kidney enriched PDZ protein) expression is limited to kidney and gastrointestinal tissues, and at subcellular levels, it can be found in cellular membranes, cytoplasm as well as endosome membranes. In renal tissues, it predominantly localize to subapical compartment of proximal tubular cells, whereas, in intestinal epithelial cells, it localizes to ileal brush border as well as in a juxtanuclear compartment. Compared to the other NHERF family members, IKEPP has few known binding partners including guanylyl cyclase C, MAP17, NHE itself, type IIa Na-Pi co-transporter, MDR protein 2, OCT family members (OCT3, OCTN1 and OCTN2), CFTR and wild-type Cl/HCO3 exchanger down-regulated in adenoma (DRA). IKEPP acts as a regulatory protein which associates with GUCY2C and negatively modulates its heat-stable enterotoxin-mediated activation. It triggers SLC9A3 activity in the presence of elevated calcium ions through IKEPP-interactions with C-terminal region of SLC34A1, GUCY2C and SLC9A3.
