Background

The type I transmembrane glycoprotein EpCAM consists of two epidermal growth factor-like extracellular domains, a cysteine-poor region, a transmembrane domain, and a short cytoplasmic tail. EpCAM is encoded by the GA733-2 gene located on the long arm of chromosome 4. EpCAM has been described by various names, including those associated with monoclonal antibodies specific for the cell surface antigen (MH99, AUA1, MOC31, 323/A3, kS1/4, GA733, and HEA125) and cDNA clones used to define the antigen kS 1/4, EGP, EGP40, and GA733-2.

PVDF membrane was probed with 0.5 µg/mL of Goat Anti-Human/Mouse E‑Cadherin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF748) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for E‑Cadherin at approximately 110 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Immunocytochemistry/Immunofluorescence: MUC-1 Antibody (SM3) [NB120-22711] - MCF7 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton-X100. The cells were incubated with anti-MUC1 [SM3] at 5 ug/ml overnight at 4C and detected with an anti-mouse IgG Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.
Immunohistochemistry-Paraffin: MUC-1 Antibody (SM3) [NB120-22711] - IHC analysis of formalin fixed paraffin embedded tissue section of human breast cancer xenograft using MUC-1 antibody clone SM3 at 1:10 dilution. The xenograft section depicted a very specific and intense signal in the periphery of the cancer cells. The necrotic cells also developed a strong immune-positivity while the tumor stroma as well as the nuclei of cells were negative for immunostaining.

  1. I'm looking for EpCAM antibody for rat. Do you have any antibody which can use FACS?
    • We unfortunately do not have any EpCAM antibodies validated in FACS for detection of the rat protein. I am sorry for any inconvenience. I do have three products to EpCAM that can detect the rat protein. These products have been validated to detect the native rat protein, so they may work for FACS however they have not yet been tested in that particular application. If you would like to try them in your experiment you would qualify for our Innovator's Reward Program. This program was designed to help scientists with the cost of their antibodies in return for valuable information. If you decide to test an antibody in a previously untested species or application we will issue you a credit for the purchase price of the antibody in exchange for a review of your experiment using our product.
