To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005).

Western Blot: Proteinase 3/Myeloblastin/PRTN3 Antibody [NBP1-25966] - Human PMN (peripheral blood mononuclear cells isolated from buffycoat; denatured, reduced) using Rabbit antibody to c-terminal region of Pr3 (Wegener autoantigen): whole serum at 1: 500 dilution; blocked with 1% LFDM for 15 minutes at room temperature with shake, primary antibody incubated for 15 minutes at room temperature, washed 3 times with PBST, 5 minutes each. Secondary antibody was also incubated for 15 minutes at room temperature.
Immunocytochemistry/Immunofluorescence: Proteinase 3/Myeloblastin/PRTN3 Antibody [NBP1-25966] - Human PBMC were isolated and adjusted to 106 cells. Cells were fixed with 2% formaldehyde for 10min at 37C. Washed twice with PBS before cytospin the cells onto microscope slides. Cells were blocked with PBS containing 1%BSA for 20min at RT. Excess blocking solution was removed and cells were incubated with Rabbit Ab to c-terminal region of Pr3 (Wegener autoantigen): whole serum for 30min at RT (diluted 1:100 in the blocking buffer). Washed 3X with PBS and incubated with anti-Rabbit Alexa 586 for further 30min. Washed as before and nuclear counterstained with DAPI. Neutrophils and Monocytes, known to have PR3 are intensely stained by the Rabbit Ab to c-terminal region of Pr3 (Wegener autoantigen): whole serum.

Western blot of primary calvarial osteoblast and skin fibroblast lysates from WT and Sc65KO 3 day-old mice (N = 2) showing significantly decreased levels of LH1 protein in Sc65KO samples. Western Blot: Plasminogen Antibody [NB600-930] - Detection of Plasminogen under reducing (R) and non-reducing (NR) conditions. Reduced samples of purified target proteins contained 4% BME and were boiled for 5 minutes. Samples of 1ug of protein per lane were run by SDS-PAGE. Protein was transferred to nitrocellulose and probed with 1:3000 dilution of primary antibody. Detection shown was using Dylight 649 conjugated Donkey anti goat 1 hr RT.
Western Blot: Plasminogen Antibody [NB600-930] - Lane 1: Plasminogen. Lane 2: None. Load: 50 ng per lane. Primary antibody: Plasminogen primary antibody at 1:1,000 overnight at 4C. Secondary antibody: Peroxidase goat secondary antibody at 1:40,000 for 60 min at RT. Blocking: incubated with blocking buffer for 30 min at RT. Predicted/Observed size: 91 kDa, 91 kDa for Plasminogen. Other band(s): None.

Western blot shows lysates of HeLa human cervical epithelial carcinoma parental cell line and Catalase knockout HeLa cell line (KO). PVDF membrane was probed with 0.5 µg/mL of Goat Anti-Human/Mouse/Rat Catalase Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3398) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017).
