Western Blot: Neutrophil Elastase/ELA2 Antibody (950334) - Azide and BSA Free [MAB9167] - Neutrophil Elastase/ELA2 Antibody (950334) [MAB9167] - Analysis of human bone marrow tissue lysate with 0.5 ug/ml of hELA-2 Monoclonal Antibody followed by HRP-conjugated Anti-Mouse IgG Antibody. A specific band was detected for hELA-2 at approximately 25-30 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Immunocytochemistry: Neutrophil Elastase/ELA2 Antibody (950334) - Azide Free [MAB9167] - Neutrophil Elastase/ELA2 was detected in immersion fixed THP-1 cell using Neutrophil Elastase/ELA2 Monoclonal Antibody at 8 ug/ml dilution. Cells were stained using Donkey anti-Mouse IgG Secondary Antibody [NL557] (red) and counterstained with DAPI (blue). Specific staining was localized to cytoplasmic.

Immunohistochemistry: Neutrophil Elastase/ELA2 Antibody (950334) [MAB9167] - ELA-2 was detected in immersion fixed paraffin-embedded sections of human lymphoma using ELA-2 Monoclonal Antibody at 5 ug/ml. Tissue was stained using Mouse IgG VisUCyte HRP Polymer Antibody (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm in neutrophils .

Flow Cytometry: Neutrophil Elastase/ELA2 Antibody (950334) - Azide Free [MAB9167] - THP-1 human acute monocytic leukemia cell line treated with 3 uM monensin for 3 hours was stained with Mouse Anti-Human Neutrophil Elastase/ELA2 Monoclonal Antibody (Catalog # MAB9167, filled histogram) or isotype control antibody (Catalog # MAB002, open histogram), followed by Phycoerythrin-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # F0102B). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005).

Background

The ELANE gene (commonly known as neutrophil elastase) encodes a 267 amino acid long, 28 kDA neutrophil elastase protein that functions in regulation of natural killer cells, monocytes, and graulocytes. ELANE is involved in activation of proMMP8, cell adhesion cell-matrix glycoconjugates, selected targets of C/EBPbeta, degradation of the extracellular matrix and collagen, and amb2 integrin signaling. It interacts with various genes GZMB, LRP1, SERPINF2, SERPING1, and SERPINA1. Elane has been linked to cystic fibrosis, asthma, pneumonia, alzheimer's disease, neutropenia, cutis laxia, leg ulcers, wegener's granulomatosis, bronchitis, adult respiratory distress syndrome, and vascular diseases.

• Comments: IHC. Single staining of neutrophil elastase (NE) in magenta on FFPE section of human liver cancer. Primary Ab is MAB9167 from Novus Biologicals. Secondary Ab is donkey anti-mouse Dylight 755. The cells exhibit a morphology that correspond to neutrophils with segmented nuclei.
