Similarly to my findings when probing against actin, in multiple species (mouse and dog), double banding appears at in tissues dense with actin. I've found that, when probing against actin, decreasing primary antibody concentration in these samples reduces, and can eliminate the non-specific band. Took a lot of troubleshooting to find the sweet spot, and I have yet to go through that process with this alpha actinin antibody.
I have found that anti-actin and actinin antibodies tend to produce a double band, especially in tissues where actin is present in high amounts. I've found that decreasing primary antibody concentration when probing for actin (1:10000), reduces this background band, but have yet to test that for this antibody.
