Western Blot: Ago2/eIF2C2 Antibody (JF0992) [NBP2-67121] - Analysis of Argonaute 2 on different lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody ( 1/500) was used in 5% BSA at room temperature for 2 hours. Immunohistochemistry-Paraffin: Ago2/eIF2C2 Antibody (JF0992) [NBP2-67121] - Analysis of paraffin-embedded human kidney tissue with Rabbit anti-Argo2/elF2C2 antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Flow Cytometry: Ago2/eIF2C2 Antibody (JF0992) [NBP2-67121] - Analysis of Hela cells with Argonaute 2 antibody at 1/50 dilution (red) compared with an unlabelled control (cells without incubation with primary antibody; black). Alexa Fluor 488-conjugated goat anti rabbit IgG was used as the secondary antibody.

Background

Ago2 / eIF2C2 encodes a member of the Argonaute family of proteins which play a role in RNA interference. The encoded protein is highly basic, and contains a PAZ domain and a PIWI domain. It may interact with dicer1 and play a role in short-interfering-RNA-mediated gene silencing.
