Flow Cytometry: BLIMP1/PRDM1 Antibody (3H2-E8) - BSA Free [NB600-235] - An intracellular stain was performed on A431 cells with BLIMP1/PRDM1 Antibody (3H2-E8) NB600-235 (blue) and a matched isotype control MAB002 (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (84540, Thermo Fisher).

Immunocytochemistry/Immunofluorescence: BLIMP1/PRDM1 Antibody (3H2-E8) [NB600-235] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton-X100. The cells were incubated with anti-BLIMP1/PRDM1 (3H2-E8) conjugated to DyLight 550 [NB600-235R] at 20ug/ml for 1 hour at room temperature. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by mouse F(ab)2 IgG (H+L) APC-conjugated secondary antibody (F0101B, R&D Systems).

The cells were incubated with anti-PCNA Antibody (PC10) NB500-106 at 1 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Proteins were visualized using a goat anti-mouse secondary conjugated to HRP and a chemiluminescence detection system.

Flow Cytometry: c-Myc Antibody (9E10) [NB600-302] - An intracellular stain was performed on U-937 cells with c-Myc Antibody (9E10) NB600-302AF488 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 10 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 488.

Immunocytochemistry/Immunofluorescence: EGLN3/PHD3 Antibody [NB100-303] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-HIF Prolyl Hydroxylase 3 (NB100-3030) at 1:200 overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at 1:500. Alpha tubulin, DM1A (NB100-690) was used as a co-stain at 1:1000 and detected with an anti-mouse DyLight 550 (Red) at 1:500. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunohistochemistry: EGLN3/PHD3 Antibody [NB100-303] - PHD3 was detected in immersion fixed paraffin-embedded sections of human heart using Rabbit Anti-Human EGLN3 polyclonal Antibody (NB100-303) at 5 ug/mL for 1 hour at room temperature followed by incubation with the Anti-Rabbit IgG VisUCyte(TM) HRP Polymer Antibody (VC003). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to nuclei.

Western blot shows lysates of HT-29 human colon adenocarcinoma cell line, SW480 human colorectal adenocarcinoma cell line, and HCT-116 human colorectal carcinoma cell line. PVDF membrane was probed with 0.5 µg/mL of Goat Anti-Human KLF4 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3640) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (HAF109). A specific band was detected for KLF4 at approximately 60 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 5.

KLF4 was detected in immersion fixed BG01V human embryonic stem cells using 10 µg/mL Goat Anti-Human KLF4 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3640) for 3 hours at room temperature. Cells were stained with the NorthernLights™ 557-conjugated Anti-Goat IgG Secondary Antibody (red; NL001) and counterstained with DAPI (blue). BG01V human embryonic stem cells were fixed using formaldehyde, resuspended in lysis buffer, and sonicated to shear chromatin. KLF4/DNA complexes were immunoprecipitated using 5 μg Goat Anti-Human KLF4 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3640) or control antibody (Catalog # AB-108-C) for 15 minutes in an ultrasonic bath, followed by Biotinylated Anti-Goat IgG Secondary Antibody (Catalog # BAF109). Immunocomplexes were captured using 50 μL of MagCellect Streptavidin Ferrofluid (Catalog # MAG999) and DNA was purified using chelating resin solution. The B2R promoter was detected by standard PCR.

KLF4 was detected in immersion fixed paraffin-embedded sections of human colon using 15 µg/mL Goat Anti-Human KLF4 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3640) overnight at 4 °C. Tissue was stained with the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). A specific band was detected for HIF-1a/HIF1A at approximately 120 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Mouse primary kidney cells treated with 150 μM CoCl2 for overnight were fixed using formaldehyde, resuspended in lysis buffer, and sonicated to shear chromatin. An intracellular stain was performed on A431 human skin carcinoma cell line using Rabbit anti-DEC1 Affinity Purified Polyclonal Antibody conjugated to Alexa Fluor® 647 (Catalog # NB100-1800AF647, blue histogram) or matched control antibody (Catalog # NBP2-24981AF647, orange histogram) at 2.5 µg/mL for 30 minutes at RT.
