Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (84540, Thermo Fisher).

Western Blot: Sodium Potassium ATPase Alpha 1 Antibody (464.6) [NB300-146] - Analysis detecting Na, K-ATPase (alpha) in porcine proximal tubule protein.

Immunocytochemistry/Immunofluorescence: Sodium Potassium ATPase Alpha 1 Antibody (464.6) [NB300-146] - Hek293 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-Sodium Potassium ATPase Alpha 1 (464.6) at 10 ug/ml overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunocytochemistry/Immunofluorescence: Sodium Potassium ATPase Alpha 1 Antibody (464.6) [NB300-146] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-Sodium Potassium ATPase Alpha 1 (464.6) at 10 ug/ml overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunocytochemistry/Immunofluorescence: Sodium Potassium ATPase Alpha 1 Antibody (464.6) [NB300-146] - Detection of ATPA1 (Green) in HepG2 cells using NB300-146. Nuclei (Blue) were counterstained using Hoechst 33258.

Western Blot: Sodium Potassium ATPase Alpha 1 Antibody (464.6) [NB300-146] - SDS–PAGE western blot validation of the increased abundance of Annexin V, Na/K ATPase & alpha -tubulin in the EVs derived from TIS Cal51 cells compared with EVs derived from non-senescent non-treated controls. EVs isolated from TIS & control cells were immunoblotted to validate the protein expression of annexin V, Na/K ATPase & alpha -tubulin from the MS data. CD63 & HSP70 were used as loading controls. Densitometry was completed using Image J software (Fiji). All values are expressed as the mean of three independent experiments±s.e.m. The limit of ±1 skewness was set as normal distribution. Skewed data were transformed to Y=Log(Y) to fit a normal distribution. Results were analysed using the Student’s t-test with significant differences having a *P⩽0.05, **P⩽0.01, ***P⩽0.001. Cells were stained using DyLight 488-conjugated Anti-Rabbit IgG (H+L) Cross-Absorbed Secondary Antibody (green) and counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.

Flow Cytometry: OCT4 Antibody [NB100-2379] - An intracellular stain was performed on Jurkat with NB100-2379 and a matched isotype control. Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody.

Immunocytochemistry/Immunofluorescence: OCT4 Antibody [NB100-2379] - Ntera2 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti- NB100-2379 at 1 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Western Blot: Park7/DJ-1 Antibody [NB300-270] - Cells were transfected with the pCMV6-ENTRY control (lane 1) or pCMV6-ENTRY PARK7 cDNA (lane 2) for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-Park7(DJ-1).

Analysis of a FFPE tissue section of mouse brain using 1:200 dilution of Park7/DJ-1 antibody. The staining was developed using HRP labeled anti-rabbit secondary antibody and DAB reagent, and nuclei of cells were counter-stained with hematoxylin.

The effect of fibrotic-ECM and tetrac on DF cell proliferation and death and on the levels of MF biomarkers, alpha v beta 3, miRNA-2,1 and D3: DF (100,000 cells/well) were cultured on normal (−TGF beta (TGFb in the graphs)) or fibrotic-ECM (+TGF beta ) with/without 0.5 μM tetrac or its solvent (DMSO-KOH propylene glycol) for 48 h. Then, cells were harvested, and the following was undertaken: (1) the cells were counted using a counter and trypan-blue; (2) the proteins were extracted from the cells, and protein levels were measured by Western blot; and (3) the RNA was extracted from the cells, and the level of miRNA-21 was evaluated using qPCR. Immunocytochemistry/Immunofluorescence: Calnexin Antibody [NB100-1974] - Rat FR cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.05% Triton X-100 in PBS for 5 minutes. The cells were incubated with (NB100-1974) at 1 ug/ml overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Western Blot: Calnexin Antibody [NB100-1974] - Identification of plasma exosomes and differentially expressed exosomal miRNAs. Confirmation of the exosomes markers with Western blotting indicated the presence of CD63 but the absence of calnexin in exosomes. $Western Blot: Calnexin Antibody - BSA Free [NB100-1974] - Immunoblot analysis of livers from control & L-Chrebp−/− mice subjected to fasting & refeeding with a high-sucrose diet. Littermate control & L-Chrebp−/− mice (same as those described in supplemental Tables S2A & S2B) were subjected to fasting & refeeding. The nonfasted (N) groups were fed chow diet ad libitum. The fasted (F) group was fasted 12 h, & the refed (R) group was fasted for 12 h & then refed with 60% (w/w) high-sucrose diet for 12 h prior to study. Liver whole-cell lysates & membrane fractions were prepared individually, & equal amounts of protein from each mouse of the same group (four per group) were pooled. Aliquots (40 μg for whole-cell lysates & 30 μg for membrane fractions) of the pooled protein were subjected to SDS-PAGE & immunoblot analysis. Immunoblot analysis of Insig-1 & Insig-2 were carried out using membrane fractions. Whole-cell lysates were used to detect other proteins. Immunocytochemistry/Immunofluorescence: DUOX2 Antibody [NB110-61576] - A431 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton-X100. The cells were incubated with anti-DUOX at 5 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Western Blot: DUOX2 Antibody [NB110-61576] - Generation and validation of NOX4 knockout (KO) HeLa cell lines. NOX4 knockout did not influence NOX1, NOX2, NOX3, NOX5, DUOX1 and DUOX2 levels. Protein levels of NOX1, NOX2, NOX3, NOX5, DUOX1 and DUOX2 in parental HeLa cells and three clones of NOX4 knockout cells. The antibody developed a strong cytoplasmic staining in the outer layer of stratum spinosum and the cells of stratum granulosum/ granular layer of skin epidermis. The endothelial cells in the dermis of the section also showed positivity for SLC31A1/CTR1.

The cancer cells developed moderate to strong positivity for CTR1/SLC31A1 protein.
