Application Notes
This Sodium Potassium ATPase (464.6) antibody is useful for Immunohistochemistry on paraffin-embedded sections, Immunocytochemistry/Immunofluorescence, Flow Cytometry and Western Blot. By Western Blot, a distinct band at 112 kDa is seen. Do not boil the sample prior to loading on the gel for Western Blot (60 degrees Celsius appears to work fine). The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors. Use in Immunohistochemistry-Frozen reported in scientific literature (PMID 26941236). Publication with sheep shown however results may vary. Use in immunprecipitation reported in scientific literature (PMID 27748972).

Background

Sodium Potassium ATPase alpha 1 (ATP1A1) belongs to the P-type ATPase family and is the catalytic component of the active enzyme, which catalyzes the hydrolysis of ATP coupled with exchange of sodium (Na+) and potassium (K+ ) ions across the cellular plasma membranes. This action creates an electrochemical gradient of Na+ and K+ ions, providing the energy for active transport of various nutrients. Its phosphorylation at Tyr-10 position alters the pumping activity whereas dephosphorylation mediated by PP2A (protein phosphatase 2A) following increases in intracellular Na+, leads to catalytic activity. Because the plasma membrane is highly permeable to water, it is the concentration of ions across this membrane that is critical for maintaining an adequate cell volume and sodium potassium ATPase is key player in this maintainance process because it provides the driving force for active Na+ as well as K+ transport into/out of the cell, with water following isosmotically. This pump is consists of equimolar ratios of two main subunits, the catalytic alpha subunit (which contains the binding sites for the mentioned cations, ATP, and cardiotonic steroid inhibitors) and the regulatory beta polypeptides (which is essential for the normal activity of the enzyme as well as involved potentially in the occlusion of K+ and modulation of the enzyme's K+ /Na+ affinity).

  1. I am looking forward to pick up a plasma membrane marker. I have gastric cancer cell line and I am preparing PM out of that. To just check the quality of prep, I need a plasma membrane marker.

  2. I would like to know whether it is possible to dilute this antibody in 50% glycerol final, and keep the aliquots at -20 degrees Celsius?
    • As far as the storage conditions are concerned, since this antibody is supplied as ascites and may contain proteases, it has to be frozen immediately at -20 or -80. From a technical perspective, adding glycerol as a cryoprotectant to avoid freeze/thaw damage should be acceptable; however, we suggest to store our antibodies as recommended in the datasheet which in this case it is better to aliquot the antibody in small volumes and store it at -20 or -80. We do supply some antibodies that we recommend adding glycerol but NB300-146 has not been tested in this storage condition.
  3. Was the epitope characterized?
    • We unfortunately have not mapped the epitope or binding region for this product yet. Our lab will always include 0.1% Tween-20 in their diluent and wash buffers for ICC staining, so I would recommend using this detergent for your customers staining. If they want to try first without the detergent, that is fine as well.
