Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 700.

Flow (Intracellular): Survivin Antibody (60.11) [NB500-238] - An intracellular stain was performed on Daudi cells with Survivin Antibody (60.11)[NB500-238] (blue) and a matched isotype control, Mouse IgG2a Kappa Light Chain Isotype Control (MG2a-53) [NB600-986] (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by mouse F(ab)2 IgG (H+L) APC-conjugated secondary antibody (F0101B, R&D Systems).

$Western Blot: Survivin Antibody (60.11) - BSA Free [NB500-238] - CD133+ cells are more resistant to chemotherapeutic agents. STA-ET-8.2 cells were FACS-sorted & CD133+ & CD133- fractions treated with increasing concentrations of (A) Doxorubicin (Doxo), (B) Etoposide (Etop), (C) Vincristine (Vinc) or (D) a combination of all three drugs at high (Doxo 10 μM; Etop 10 ug/ml; Vinc 100 ng/ml), medium (Doxo 2.5 μM; 20 Etop 2.5 ug/ml; Vinc 25 ng/ml) & low (Doxo 0.5 μM; Etop 0.5 ug/ml; Vinc5 ng/ml) concentrations. Viability was assessed by MTS assay after 96 hrs (*p < 0.05) & CD133+ cells displayed increased drug resistance. (E): CD133+ & CD133- TC-71 cells are equally sensitive to Doxo, Etop, & Vinc. (F): Western blot reveals no difference in apoptosis & drug resistance protein expression between unsorted & CD133-sorted STA-ET-8.2 cell fractions. TLR4 Inhibitor Peptide Set [NBP2-26244] - TLR4/MD-2/CD14/NF-kB/SEAPorter stably transfected cells were plated in 96-well plates at 1x10^5 cells/well. After 16 h, cells were preincubated with various concentrations of inhibitor or control peptides (CP) for 1 h. Cells were then stimulated with 10 ng/ml LPS for 24 h. The secreted alkaline phosphatase (SEAP) was analyzed using the SEAPorter Assay Kit. Inhibitory effect on the LPS-mediated TLR4 activation by the VIPER peptide is shown in panel A, and IC50 of the VIPER peptide is described in panel B.

Western Blot: STING/TMEM173 Antibody [NBP2-24683] - Total protein from THP-1, HT-29, U2OS cells and human spleen was separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml STING/TMEM173 Antibody in 1% non-fat milk in TBST and detected with an anti-rabbit HRP secondary antibody using chemiluminescence.

Immunohistochemistry-Paraffin: STING/TMEM173 Antibody [NBP2-24683] - Human colon cancer tissue section using STING/TMEM173 Antibody at 1:100 dilution with detection employing HRP-conjugated secondary antibody. The signal was developed using DAB reagent and the nuclei were counterstained with hematoxylin. The antibody generated very weak cytoplasmic staining in columnar epithelial cells with a very strong signal in the secretory/goblet cells.

Immunohistochemistry-Paraffin: STING/TMEM173 Antibody [NBP2-24683] - Mouse lung tissue section using STING/TMEM173 Antibody at 1:150 dilution with detection employing HRP-conjugated secondary antibody. The signal was developed using DAB reagent and the nuclei were counterstained with hematoxylin. The antibody generated mainly a cytoplasmic staining in the bronchiolar and alveolar epithelial cells.

Flow Cytometry: STING/TMEM173 Antibody [NBP2-24683] - An intracellular stain was performed on U937 cells with NBP2-24683AF594 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 594.

Flow Cytometry: STING/TMEM173 Antibody - BSA Free [NBP2-24683] - An intracellular stain was performed on THP-1 cells with STING/TMEM173 Antibody NBP2-24683AF647 (blue) and a matched isotype control NBP2-24891 (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 647.

Immunocytochemistry/Immunofluorescence: STING/TMEM173 Antibody [NBP2-24683] - HT-29 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton-X100. The cells were incubated with STING/TMEM173 Antibody at 10 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Flow Cytometry: STING/TMEM173 Antibody [NBP2-24683] - An intracellular stain was performed on THP-1 cells with STING/TMEM173 Antibody and a matched isotype control. Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody.

Flow Cytometry: STING/TMEM173 Antibody [NBP2-24683] - An intracellular stain was performed on RH30 cells with STING/TMEM173 Antibody (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody.

<b>Genetic Strategies Validation. </b>Western blot shows lysates of THP-1 cell line and STING/TMEM173 knockout THP-1 cell line (KO). Nitrocellulose membrane was probed with STING/TMEM173 Antibody (Catalog # NBP2-24683) followed by an HRP-conjugated secondary antibody. A specific band was detected for STING/TMEM173 at approximately 41 kDa (as indicated) in the parental THP-1 cell line, but is not detectable in knockout THP-1 cell line. Primary antibody dilution used: 1/1000. The Ponceau stained transfer of the blot is shown. This experiment was conducted under reducing conditions. $PMA-treated THP-1 lysates were prepared and immunoprecipitation was performed using 2.0 µg of STING/TMEM173 Antibody (Catalog # NBP2-24683) pre-coupled to Dynabeads protein A. Immunoprecipitated STING/TMEM173 was detected with STING/TMEM173 Antibody (Catalog # NBP3-18816). For western blot, NBP3-18816 was used at 1/1000. The Ponceau stained transfer of the blot is shown. Simple Western: A20/TNFAIP3 Antibody (59A426) [NBP1-77533] - Lane view shows a specific band for A20/TNFAIP3 in 0.05 mg/ml of Jurkat lysate. This experiment was performed under reducing conditions using the 12-230 kDa separation system.

Immunohistochemistry-Paraffin: A20/TNFAIP3 Antibody (59A426) [NBP1-77533] - Human placenta probed with A20 antibody at 5 ug/ml, cytoplasmic staining of decidual cells is observed. Human tissue TMA was used for this test. Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10 mM sodium citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min.

Immunocytochemistry/Immunofluorescence: A20/TNFAIP3 Antibody (59A426) [NBP1-77533] - HepG2 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton-X100. The cells were incubated with anti-A20/TNFAIP3 (59A426) at 10 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective

The cells were incubated with anti-A20/TNFAIP3 Antibody [59A426] NBP1-77533 at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Flow Cytometry: A20/TNFAIP3 Antibody (59A426) [NBP1-77533] - An intracellular stain was performed on RH-30 cells with A20/TNFAIP3 Antibody (59A426) NBP1-77533PE (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Phycoerythrin.

A20/TNFAIP3 (59A426) was detected in immersion fixed Caco-2 human colorectal adenocarcinoma cell line using Mouse anti-A20/TNFAIP3 (59A426) Protein G Purified Monoclonal Antibody conjugated to Alexa Fluor® 647 (Catalog # NBP1-77533AF647) (light blue) at 10 µg/mL overnight at 4C. Cells were counterstained with DAPI (dark blue). Cells were imaged using a 40X objective.

Immunohistochemistry: CCR7 Antibody [NB100-712] - M1 expression pattern changed in trabecular bone. Immunohistochemical staining was performed in sagittal serial sections of medial condyles from 6-week-old rats within the epiphyseal region. The region with the solid line indicates areas of interest to be enlarged in the following image. Representative staining with the anticluster of differentiation 68 (CD68) antibody demonstrated that CD68+ cells were either attached (black arrows) or unattached (white arrows) to the bone surfaces with a variety of sizes & shapes (a, b). TRAP+ cells showed multinucleated morphology (black arrows) which is similar to the CD68+ cells (c, d). Inducible nitric oxide synthase-positive (iNOS+) & C-C chemokine receptor type 7-positive (CCR7+) cells (black arrow) were attached to the bone surfaces with an elongated morphology (e, f, i, j). Arginase1+ & cluster of differentiation 163-positive (CD163+) cells (white arrow) represented the predominant population in the reticular connective tissue with similar expression patterns as in the cortical bone (g, h, k, l). No positive staining was found in isotype controls (data not shown). Most of the M1-labeled cells were found attached to bone surfaces instead of M2-labeled cells (m). Slides were stained for PSMA, EpCAM, sLex, & CXCR4 using the protocol as described in Figure S1. MDA, PC3, & KG1 cells were simultaneously stained as a control for the following markers: PSMA= Magenta, EpCAM= Yellow, HECA-452= Green, CXCR4= Red. All prostate CTCs expressed CXCR4, while, sLex expression was variable. The analysis of sLex intensity is shown in Figure 4. Immunocytochemistry/Immunofluorescence: TNF-alpha Antibody - Azide Free [NB600-587] - LMP2 knockdown by shRNA reduced the expression of TNF-alpha and IL-1beta after MCAO. Immunofluorescence labeling revealed that both TNF-alpha and IL-1beta immunoreactivity was significantly upregulated in the ipsilateral ischemic hemisphere compared with sham-operated animals. In contrast, LMP2 knockdown by shRNA significantly reduced the expression of TNF-alpha and IL-1beta. Five rats in each group were used for immunofluorescent assays (n=5). Immunohistochemistry: TNF-alpha Antibody - Azide Free [NB600-587] - Staining of formalin/PFA-fixed paraffin-embedded sections of human artery tissue sections. Sections were fixed in formaldehyde and subjected to heat mediated antigen retrieval in citrate buffer (pH 6.0). Slides were blocked for ten minutes with 1.5% serum. Primary antibody was diluted 1:100 and incubated with samples for 24 hours at 4C. HRP-conjugated goat anti-rabbit antibody was used as the secondary antibody.

Samples were formaldehyde-fixed, then blocked in 10% serum for 2 hours at 20C. The primary antibody was diluted 1:100 and incubated with the sample for 2 hours at 20C.
