![Immunohistochemistry-Paraffin: CCR7 Antibody [NB100-712] - M1 and M2 macrophages showed similar expression patterns in cortical bone. IHC staining was performed in sagittal serial sections of medial condyles from 6-week-old rats within the diaphyseal region. Cluster of differentiation 68 (CD68) was selected as a pan marker for the macrophage lineage, including monocytes, macrophages, giant cells, and osteoclasts. iNOS and C-C chemokine receptor type 7 (CCR7) were selected as M1 phenotypic markers, whereas Arginase1 and cluster of differentiation 163 (CD163) were selected as M2 phenotypic markers. Tartrate-resistant acid phosphatase (TRAP) staining was performed as described in Materials and Methods. Quantification of cell numbers according to their location revealed that most of the M1 and M2 cells were not attached to the bone surfaces. ![Immunohistochemistry: CCR7 Antibody [NB100-712] - M1 expression pattern changed in trabecular bone. Immunohistochemical staining was performed in sagittal serial sections of medial condyles from 6-week-old rats within the epiphyseal region. The region with the solid line indicates areas of interest to be enlarged in the following image. Representative staining with the anticluster of differentiation 68 (CD68) antibody demonstrated that CD68+ cells were either attached (black arrows) or unattached (white arrows) to the bone surfaces with a variety of sizes & shapes (a, b). TRAP+ cells showed multinucleated morphology (black arrows) which is similar to the CD68+ cells (c, d). Inducible nitric oxide synthase-positive (iNOS+) & C-C chemokine receptor type 7-positive (CCR7+) cells (black arrow) were attached to the bone surfaces with an elongated morphology (e, f, i, j). Arginase1+ & cluster of differentiation 163-positive (CD163+) cells (white arrow) represented the predominant population in the reticular connective tissue with similar expression patterns as in the cortical bone (g, h, k, l). No positive staining was found in isotype controls (data not shown). Most of the M1-labeled cells were found attached to bone surfaces instead of M2-labeled cells (m). ![Immunocytochemistry/Immunofluorescence: CXCR4 Antibody [NB100-74396] - Optimization of immunofluorescence staining of CXCR4 protein. MDA, PC3, KG1 cells were used for the optimization experiments. Cells were seeded onto cell-tak coated 48 mm coverslips in a 48-well plate. Cells were fixed with 2% formaldehyde for 20 min, washed with PBS. After fixation, cells were blocked with 2.0% BSA in PBS for 1 h at room temperature. All the cells were incubated with a primary antibody for anti-rabbit CXCR4 for 1 h. After washing with PBS, cells were put in respective secondary antibodies-anti-rabbit dylight 405 for 1 h. PSMA= Magenta, EpCAM= Yellow, sLex= Green, CXCR4= Red, and Merge shows all the colors. Data are presented as the mean ± SD. #Value significantly greater than the corresponding value in saline-treated normoxic mice (##P 50 CTCs/ 2.5 ml blood) was processed via ficoll density centrifugation & the PBMC fraction was collected. Immunomagnetic anti-CD45 depletion was performed on the obtained PBMCs & the remaining cells were washed, cytospunned onto the slides. Slides were stained for PSMA, EpCAM, sLex, & CXCR4 using the protocol as described in Figure S1. MDA, PC3, & KG1 cells were simultaneously stained as a control for the following markers: PSMA= Magenta, EpCAM= Yellow, HECA-452= Green, CXCR4= Red. ![<b>Biological Strategies Validation. </b>Immunohistochemistry-Paraffin: CCL2/MCP1 Antibody [NBP1-07035] - IHC analysis of a formalin-fixed paraffin-embedded (FFPE) human breast carcinoma tissue section using 1:1000 dilution of CCL2/MCP1 antibody (NBP1-07035) on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) with 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching using peroxide block. The sections were incubated with primary antibody for 30 minutes. Bond Polymer Refine Detection (Leica Biosystems) and DAB were used for signal detection which followed counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) were performed using Aperio AT2 (Leica Biosystems). This antibody generated a diffused cytoplasmic staining of CCL2 antigen in the cancer cells, stromal cells as well as the endothelial cells. The stroma itself showed a weak immunopositivity for CCL2.
