The cisplatin resistant CAL27cis & Detroit‐562cis cells had increased IL‐6 mRNA & IL‐6 protein production compared to parental cells by qRT‐PCR & ELISA (A, B). CAL27cis & Detroit‐562cis were cultured with IL‐6 (10 ng/mL, human recombinant IL‐6, R&D Systems, Minneapolis, USA) for 48 h (C). An anti‐human IL‐6 polyclonal antibody (R&D Systems, Minneapolis, USA) was used to neutralize the biological activities of IL‐6 (E, F). To inhibit signal transduction, CAL27cis & Detroit‐562cis cells were pre‐incubated for 1 h with STAT3 inhibitor V (Sigma‐Aldrich, Germany, 500 nmol/L), & then incubated with recombinant human IL‐6 (10 ng/mL) for 24 h before analysis. The cells were incubated with anti CD68 (FA-11) [NBP2-33337] at a 1:100 dilution overnight at 4C and detected with an anti-rat DyLight 488 (Green) at a 1:500 dilution. ![Immunohistochemistry: CCR7 Antibody [NB100-712] - M1 expression pattern changed in trabecular bone. Immunohistochemical staining was performed in sagittal serial sections of medial condyles from 6-week-old rats within the epiphyseal region. The region with the solid line indicates areas of interest to be enlarged in the following image. Representative staining with the anticluster of differentiation 68 (CD68) antibody demonstrated that CD68+ cells were either attached (black arrows) or unattached (white arrows) to the bone surfaces with a variety of sizes & shapes (a, b). TRAP+ cells showed multinucleated morphology (black arrows) which is similar to the CD68+ cells (c, d). Inducible nitric oxide synthase-positive (iNOS+) & C-C chemokine receptor type 7-positive (CCR7+) cells (black arrow) were attached to the bone surfaces with an elongated morphology (e, f, i, j). Arginase1+ & cluster of differentiation 163-positive (CD163+) cells (white arrow) represented the predominant population in the reticular connective tissue with similar expression patterns as in the cortical bone (g, h, k, l). No positive staining was found in isotype controls (data not shown). Most of the M1-labeled cells were found attached to bone surfaces instead of M2-labeled cells (m). ![Immunocytochemistry/Immunofluorescence: LAMP-2/CD107b Antibody (H4B4) [NBP2-22217] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-LAMP-2/CD107b [NBP2-22217] at a 1:100 dilution overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue).
