![Immunocytochemistry/ Immunofluorescence: CD4 Antibody - BSA Free [NBP1-19371] - Rats with mild liver damage show immune cells infiltration in cerebellum at 4 weeks, which is prevented by rifaximin. Analysis of immune cells infiltration in cerebellum was performed at 4 weeks by immunohistochemistry using antibodies against (A) CD4, a marker of T lymphocytes (F(3,8) = 4.511, p < 0.01) & (B) IBA1, a marker of meningeal & perivascular macrophages (K-W(4,10) = 27.76, p < 0.0001). (C) Double immunofluorescence with anti-CD4 & anti-CX3CR1 as markers of autoreactive CD4+CD28− T lymphocytes was performed & quantified (K-W(4,5) = 8.711, p < 0.05). (D) Double immunofluorescence with anti-CD4 & anti-CCR6 as marker of Th17 lymphocytes was performed & quantified (K-W(4,7) = 8.858, p < 0.05). Number of animals in each group was added under group names. One-way ANOVA with Tukey’s test (CD4+) & nonparametric Kruskal–Wallis (K-W statistic) with Dunn’s test (Iba1, CD4+/CX3CR1+ & CD4+/CCR6+) was performed to compare all groups. Values significantly different from control rats are indicated by asterisks & from CCl4 rats by a. * p < 0.05, a p < 0.05. The cisplatin resistant CAL27cis & Detroit‐562cis cells had increased IL‐6 mRNA & IL‐6 protein production compared to parental cells by qRT‐PCR & ELISA (A, B). CAL27cis & Detroit‐562cis were cultured with IL‐6 (10 ng/mL, human recombinant IL‐6, R&D Systems, Minneapolis, USA) for 48 h (C). An anti‐human IL‐6 polyclonal antibody (R&D Systems, Minneapolis, USA) was used to neutralize the biological activities of IL‐6 (E, F). To inhibit signal transduction, CAL27cis & Detroit‐562cis cells were pre‐incubated for 1 h with STAT3 inhibitor V (Sigma‐Aldrich, Germany, 500 nmol/L), & then incubated with recombinant human IL‐6 (10 ng/mL) for 24 h before analysis. ![Dual RNAscope ISH-IHC: HLA-DR Antibody (L243) [NB100-77855] - Formalin-fixed paraffin-embedded tissue sections of human metastatic tonsil were probed for HLA-DR mRNA (ACD RNAScope probe, catalog # 475891; Fast Red chromogen, ACD catalog # 322500). Adjacent tissue section was processes for immunohistochemistry using mouse monoclonal (Novus catalog # NB100-77855) at 1:50 dilution for 1 hour at room temperature followed by incubation with the anti-mouse IgG VisUCyte HRP Polymer Antibody (Catalog # VC001) and DAB chromogen (yellow-brown). ![Immunocytochemistry/Immunofluorescence: CD68/SR-D1 Antibody (FA-11) [NBP2-33337] - Wehi-3 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti CD68 (FA-11) [NBP2-33337] at a 1:100 dilution overnight at 4C and detected with an anti-rat DyLight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue).
