![<b>Genetic Strategies Validation. </b>Western Blot: Bcl-2 Antibody [NB100-56098] - EA reduced MPP+-induced dopaminergic neuronal apoptosis by increasing BDNF (brain-derived neurotrophic factor) expression and further Akt phosphorylation in the rat substantia nigra. Eight days after MPP+ administration, our Western blot results (MAB7566) show that MPP+ treatment reduced tyrosine hydroxylase and Bcl-2 expression in the ipsilateral side of the rat substantia nigra (SN), but not in the contralateral side. ![<b>Biological Strategies Validation. </b>Immunohistochemistry-Paraffin: CCL2/MCP1 Antibody [NBP1-07035] - IHC analysis of a formalin-fixed paraffin-embedded (FFPE) human breast carcinoma tissue section using 1:1000 dilution of CCL2/MCP1 antibody (NBP1-07035) on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) with 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching using peroxide block. The sections were incubated with primary antibody for 30 minutes. Bond Polymer Refine Detection (Leica Biosystems) and DAB were used for signal detection which followed counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) were performed using Aperio AT2 (Leica Biosystems). This antibody generated a diffused cytoplasmic staining of CCL2 antigen in the cancer cells, stromal cells as well as the endothelial cells. The stroma itself showed a weak immunopositivity for CCL2. ![Western Blot: TERT Antibody (2C4) [NB100-317] - IFI16 inhibits c-Myc-stimulated transcription & hTERT expression in HeLa cells.(A) Total protein extracts prepared from HeLa cells infected with control retrovirus (lane 1) or a virus encoding IFI16 protein (lane 2) were subjected to immunoblotting using antibodies specific to the indicated proteins. (B) Sub-confluent cultures of HeLa cells were transfected with pMyc-TA-luc reporter plasmid (1.0 µg) along with a second pRL-TK reporter plasmid (0.2 µg) & an empty plasmid (pCMV; column 1), a plasmid encoding c-Myc (column 2 & 4), a plasmid encoding IFI16 (column 3), or both plasmids encoding c-Myc & increasing amounts of the plasmid encoding IFI16 protein (column 5 & 6). After 44–48 h of transfections, cells were lysed & the lysates were analyzed for dual luciferase activity. Normalized relative luciferase activity in control cells is indicated as 1.0. ![Immunohistochemistry: ATM Antibody (5C2) [NB100-220] - Limited role of Atm in p53-mediated tumor suppression & DNA damage response in chemically-induced fibrosarcomas.A. Mice of the indicated genotypes, wt (n = 15), p53-super (n = 16), Atm-null (n = 9), p53-super/Atm-null (n = 13) & Arf-null (n = 12), were injected intramuscularly with 3-methyl-cholanthrene (3MC) & tumour development was monitored. Kaplan-Meier tumour-free curves were obtained & statistical significant differences (logrank test) were found for wt vs. p53-super (p<0.005), Atm-null vs. p53-super/Atm-null (p<0.001) & Arf-null vs. wt (p<0.0001). No significant differences were found for wt vs. Atm-null (p = 0.11), or p53-super vs. p53-super/Atm-null (p = 0.18). B. Quantification of the persistent DNA damage response ( gamma H2AX), proliferation (Ki67), & apoptosis (activated caspase-3) in 3MC-fibrosarcomas generated in mice of the indicated genotypes.
